摘要 目的:探讨高血压时卵巢细胞凋亡的调控机制。方法:以日本引进DahlS高血压大鼠为研究对象,应用流式细胞仪、生化等技术,对DahlS高血压大鼠卵巢细胞凋亡与Bcl-2、iNOS及NO等相关因素进行研究。结果:(1)随高血压病情加重,卵巢细胞凋亡率逐渐上升,与之伴随Bcl-2表达率逐渐下降,iNOS表达率逐渐升高,实验组与对照组比较差别显著,P<0.05。(2)11周龄实验组血清NO含量较对照组降低,两者比较差别显著,P<0.01;而随高血压病情加重,实验组NO含量逐渐增高,反而高于对照组,两组比较差异有统计学意义,P<0.05。丙二醛(MDA)随高血压病情加重逐渐增高,实验组与对照组比较差别显著,P<0.05。结论:随血压升高,卵巢细胞凋亡加重与Bcl-2调控及iNOS、NO直接作用有关。 Objectives To investigate the characteristics and mechanism of regulation of hypertension induced ovary cell apoptosis, and to study the apoptosis and its related factors of ovarian cell in DahlS rats. Methods;Sixty DahlS hytertensive rats were randomly divided into 6 groups, 3 of which were experimental groups and given food with 8% salt, and the other 3 groups were control groups and fed with normal food. The relations between Bcl-2, NO,iNOS, MDA and the apoptosis of ovarian cell were studied by using flowcytometry, biochemical assay techniques. Results:The apoptosis rate of ovarian cell increased gradually with development of hypertension, while the expression of Bcl-2 decreased and the expression of iNOS increased, the difference between the experimental and the control groups was significant (.P<0. 01). The level of NO, MDA also increased significantly. Conclusion:Hypertension can aggravate the apoptosis of ovarian cell, it was related with regulation of Bcl-2 and direct action of NO and iNOS.
