摘要
比较了常用的三种提取天花粉蛋白(Tichosanthin,TCS)方法的提取效果,通过SDS-PAGE分析了大苞栝楼籽的蛋白组成和分子质量分布,以及大苞括楼的根、叶、茎和愈伤组织中蛋白组成的异同,同时分析了各个器官中26ku蛋白的氨基酸组成。结果表明,Wong的方法较好;栝楼和大苞栝楼的图谱符合程度为56.3%,图谱特征系数为43.7%;大苞栝楼的各个器官的蛋白组成差异较大,其中多数为低分子质量蛋白,具有很多共同的谱带,而且,根、叶、茎和种子中都具有与TCS和核糖体失活蛋白TCK电泳行为相似的蛋白,氨基酸分析也证明了它们组成上的一致性。从而证明大苞栝楼具有潜在的药用价值。
This paper presents the comparison of the results of three commonly used methods for trichosanthin (TCS) extraction, and the protein composition and their molecular weight (Mr) ranges were determined applying SDS-polyac-rylamide gel electrophoresis, the protein composition in the organs of stem, leaf, root tuber, seed and callus of Tricho-santhes bracteata Voigt. And the amino acid compositions of the 26ku protein present in the samples were analyzed to determine the homogeneity among them. The results showed that the Wong method was the best of the three tested, and the electrophoretograms of the seed samples of the two species showed a 56.3 percentage of pattern homology, 43.7 percentage of pattern heterology. An apparent difference between the protein compositions of the samples of Trichosan-thes bracteata Voigt was observed, most of which are of low Mr.Also, many bands of same Mr were observed in all of the electrophoretograms. Proteins showing similar electrophoresis behaviors to those of TCS and trichokirin (TCK) were observed and amino acid compositions confirmed their homogeneity, therefore showing Trichosanthes bracteata Voigt's pharmacies value.
