摘要 本研究采用基于气相色谱-质谱(GC-MS)的非靶向代谢组学方法,观察百草枯中毒大鼠血浆中的代谢变化及血必净注射液(XBJ)的治疗效果。通过化学计量学方法,在百草枯中毒组和对照组之间共鉴定出7种差异代谢物,其中5种代谢物质在XBJ处理后受到明显回调,改变的代谢途径包括能量代谢、氨基酸代谢、脂肪酸代谢和糖代谢。研究结果表明,基于GC-MS的代谢组学可以揭示百草枯中毒潜在的生物标志物,且XBJ可通过调节多种代谢途径发挥治疗作用。 The use of metabolomics to analyze the biological samples is critical to elucidate the pathogenesis of paraquat poisoning and assess the therapeutic effects of drugs.In this work,we used the GC-MS based non-targeted metabolomics method to observe the metabolites changes in plasma of paraquat poisoning rats and the therapeutic effect of Xuebijing injection(XBJ).Seven differential metabolites were identified in the paraquat poisoning group and the control group by chemometric methods,in which five were significantly retreated after the treatment of XBJ.The metabolism pathway of energy,amino acid,fatty acid,and saccharide was changed.These results suggest that GC-MS-based metabolomics may help find the potential biomarkers of paraquat poisoning,and XBJ plays a therapeutic role by regulating multiple metabolic pathways.
