摘要
目的研究加味当归芍药散对乳腺增生大鼠乳腺上皮细胞凋亡和Bcl-2表达的影响及作用机理。方法采用雌、孕激素联合造模制成大鼠乳腺增生模型,随机分为正常对照组、模型组、加味当归芍药散高、中、低剂量组、乳癖消对照组;TUNEL法检测计算6组动物乳腺上皮细胞凋亡指数;免疫组化S-P法检测Bcl-2表达情况。结果各治疗组与模型组比较,乳腺上皮细胞凋亡指数均明显增加(P<0.01),加味当归芍药散高剂量组和乳癖消组优于加味当归芍药散中、低剂量组(P<0.01,P<0.05);各治疗组与模型组比较,Bcl-2表达均降低(P<0.01)。结论加味当归芍药散可能通过调理肝脾、化瘀散结,促进乳腺上皮细胞凋亡并抑制Bcl-2表达达到治疗作用。
Objective To observe the effects of the supplemented Danggui Shaoyao san(SDSS) on apoptosis of mammary epithelial cells and Bcl-2 expression in hyperplasia of mammary glands rats, and ex-plore its mechanism. Methods The rat models of hyperplasia of mammary glands were established by using estrogen combined with progestogen. The rats were randomly divided into blank control group, model group,high, middle and low dose groups of SDSS and Rupixiao control group. The apoptotic index of mammary ep-ithelial cells was detected by TUNEL method, and the Bcl-2 expression was detected by Immunohistochemical S-P method. Results Compared with model group, the apoptotic index of mammary epithelial cells in each treatment group was increased(P<0.01), the index in high dose group of SDSS, Rupixiao control group was respectively higher than that of middl, low dose groups of SDSS(P<0.01, P<0.05), and the Bcl-2 expression in each treatment group was decreased(P<0.01). Conclusion SDSS has good effect on cyclomastopathy,and the mechanism is probably related with harmonizing liver-spleen, and removing stasis, promoting apoptosis of mammary epithelial cells, and inhibiting Bcl-2 expression.
