摘要
目的研究加味四君子汤体外诱导淋巴瘤Raji细胞凋亡及其作用机制。方法不同浓度的加味四君子汤作用于Raji细胞,MTT比色法分析细胞增殖情况,荧光染色法观察细胞形态学变化,流式细胞术分析细胞凋亡情况,RT-PCR检测Bax、Bcl-2 m RNA的表达。结果 1加味四君子汤对Raji细胞的增殖有较强的抑制作用;2显微镜下可见明显的细胞凋亡;3流式分析显示加味四君子汤作用Raji细胞72 h后可显著诱导细胞凋亡;4 RT-PCR显示,Bcl-2 m RNA的表达随着药物浓度增加而下降,而Bax m RNA表达增加。结论加味四君子汤能有效地诱导Raji细胞凋亡,抑制细胞增殖,其机制可能与Bax基因表达增加及Bcl-2基因表达降低有关。
Objective To study the effect of Jiawei Sijunzi decoction on Raji cells apoptosis in vitro, and explore its mechanism. Methods Raji cells were treated with Jiawei Sijunzi decoction with different concentrations, the cells proliferation was investigated by MTT assay, the morphology of the cells was observed by fluorescent staining method, the cells apoptosis was studied by flow cytometry, and the expressions of Bcl-2and Bax m RNA were measured by RT-PCR. Results Jiawei Sijunzi decoction inhibited the proliferation of Raji cells. Under the microscope, karyopyknosis was seen, chromatin margination and anachromasis were found in the cells. After giving Jiawei Sijunzi decoction for 72 hours, the apoptosis of Raji cells was significant by flow cytometry. With the increase of drug concentration, the expression of Bcl-2 m RNA was decreased, and the expression of Bax m RNA was increased. Conclusion Jiawei Sijunzi decoction can induce Raji cells apoptosis effectively, inhibit their proliferation, and the probably mechanism is related to up-regulating the expression of Bax m RNA and down-regulating the expression of Bcl-2 m RNA.
