摘要
本研究以海陆渐渗系13-1×辽棉12组配的195个单株的F2群体为作图群体,构建遗传连锁图谱,挖掘与纤维品质相关稳定的QTL,为标记辅助13-1选择育种提供依据。本研究利用SSR标记和Joinmap3.0软件构建遗传连锁图谱,并通过Ici Mapping完备区间作图法对F2及F2:3家系进行纤维品质性状QTL定位。结果显示:构建的遗传连锁图谱包含39个多态性标记、13个连锁群,该图谱总长1 174.4 c M,覆盖棉花基因组的26.7%;共检测到37个与纤维品质相关的QTL,其中伸长率2个,整齐度9个,马克隆值19个,比强度7个,分布在10个染色体上。19个有利等位基因来自海陆渐渗系13-1,18个有利等位基因来自辽棉12,发现两个稳定的QTL位点,可作为海陆渐渗系棉花纤维品质基因功能研究的候选基因。本研究筛选出的多态性标记可辅助前期选择。
In this study, a genetic linkage map was constructed by the 195 F2 population derived from the cross between the parents of Introgression Lines13-1 and Liao12 to map QTLs related with fiber qualities, and further to provide information for molecular marker-assisted breeding. The linkage map was constructed with SSR markers by JOINMAP 3.0 software. The QTLs for fiber quality properties in F2 and F2:3populations were mapped by the inclusive composite interval mapping method with Ici Mapping. The map included 39 SSR loci and 13 linkage groups, covering 1 174.4 c M and accounting for 26.7% of the cotton genome; 37 QTLs related fiber quality were identified among 10 chromosomes in F2 and F2:3population, including 2 QTLs for fiber elongation, 9 for fiber length uniformity, 19 for fiber micronaire and 7 for fiber upper half mean length. There are 19 favorable alleles originated from Introgression Lines13-1 and 18 favorable alleles originated from Liao12.Two stable QTLs could be the candidate alleles for gene function research to reveal the molecular regulation mechanism as well as for MAS breeding projects.
