摘要
植物14-3-3蛋白(GF14蛋白)是一类参与调节植物生命活动过程的小分子蛋白。本研究根据高羊茅转录组学测序获得的Fa GF14-A和Fa GF14-D基因片段,利用RACE技术得到高羊茅Fa GF14-A和Fa GF14-D基因的3'和5'c DNA序列,拼接后的Fa GF14-A基因全长为1 153 bp,编码260个氨基酸;Fa GF14-D基因全长为1 291 bp,编码266个氨基酸。生物信息学分析结果表明高羊茅Fa GF14-A和Fa GF14-D蛋白都具有典型的14-3-3蛋白结构域,包含9个保守的琢-螺旋及不保守的N-末端和C-末端,尤其与禾本科植物GF14蛋白具有较高的相似性,位于同一个进化支上,亲缘关系较近。
Plants 14-3-3 protein(GF14) is a kind of small molecules proteins participating in the process of plant life activities. Using FaGF14-A and Fa GF14-D gene fragment from tanscriptome sequencing as template. The full length c DNA of Fa GF14-A and Fa GF14-D gene from tall fescue has been identified by RACE methods. Sequence analysis shows that Fa GF14-A is 1 153 bp in length,encode proteins with 260 amino acids,Fa GF14-D is 1 291 bp in length,encode proteins with 266 amino acids. Bioinformatics analysis reveals that proteins encoded by the two genes both contain two conserved 14-3-3 domains,9 conserved alpha helix and non-conserved C-terminus,N-terminus in secondary structure,Fa GF14-A and Fa GF14-D showed a close genetic relationship with the known GF14 of gramineous plants.
