马尾松PmLFY和PmNLY基因的克隆与表达分析

摘要 本研究在前期马尾松转录组测序中获得两个LFY同源基因片段的基础上,克隆获得Pm LFY和Pm NLY基因全长,分别为1 164和1 212 bp,编码387和403个氨基酸。生物信息学分析表明,2个LFY同源基因都具有典型的FLO_LFY结构域,但Pm NLY基因缺失了XEL…XTL…XEL…拉链结构。同源性和进化分析表明,LFY和NLY基因分别单独分为一类,并且与针叶植物的亲缘关系非常近。表达分析表明,2个基因都为组成型表达,均主要参与了雌球花发育过程,并且Pm LFY还参与了球果发育。本研究为今后马尾松花发育机理研究提供理论依据。 In this study, the full-length sequence of Pm LFY and Pm NLY genes cloned from Pinus massoniana L.using RT-PCR and RACE base on floral development transcriptome sequencing. Pm LFY and Pm NLY gene cloned the full-length sequence of the complete open reading frames of both genes were 1 164 and 1 212 bp, encoding 387 and 403 amino acids, respectively. The amino acid sequence for gene bioinformatics analysis showed, Pinus massoniana have typically FLO_LFY domains for two LFY homologous genes, but Pm NLY gene deletion XEL...XTL... XEL... domains. Phylogenetic analysis showed that LFY and NLY gene into a separate category,respectively. Also did not come together for LFY and NLY genes of in the same species, and kinship conifers very close. We tested the expression levels of Pm LFY and Pm NLY using Quantitative PCR in different tissues, male cones and female cones under different developmental phases. The results showed that the two genes are constitutively expressed, the two genes main involved in the development process of the female cones and Pm LFY also involved in strobile development, maybe. This study for future development mechanism study provide theoretical basis for pinus massoniana flowers.
