利用SSR标记分析油菜菌核病抗性资源遗传多样性 被引量:2

Genetic Diversity of Rapeseed(Brassica napus) with Resistance to Sclerotinia sclerotiorum Analyzed based on SSR Markers

摘要 为掌握油菜菌核病抗性资源遗传多样性,揭示材料的遗传背景,明确材料间的亲缘关系,从而促进抗菌核病油菜育种。本研究利用40对SSR核心引物,对汉中市农科所经过茎秆接种鉴定的菌核病抗性表现较好的43份油菜材料进行遗传多样性分析,利用NTSYS-pc2.10e软件计算材料间遗传相似系数,UPGMA法进行聚类分析。研究表明,40对SSR引物共检测到291个条带,平均每个标记检测到7个条带,每个SSR位点的遗传多态性信息含量PIC在0.53~0.98之间,平均值为0.76。供试43份油菜材料遗传相似系数主要分布在0.66~0.76之间,遗传相似度较高。本研究表明油菜抗菌核病资源遗传多样性较低,亲缘关系较近,应进一步加强抗性资源筛选,丰富现有油菜遗传背景,从而促进抗菌核病油菜新品种选育。 In order to analyze the genetic diversity of rapeseed(Brassica napus) with resistance to Sclerotinia sclerotiorum, to reveal the genetic background, and to clear the materials genetic relationship, promoting the breeding process of Sclerotinia sclerotiorum resistant rapeseed, forty core SSR primers were used to analyze 43 Sclerotinia sclerotiorum resistant rapeseed materials identified with stem inoculation method by Hanzhong Institute of Agricultural Sciences. Genetic similarity coefficient was calculated by using NTSYS-pc2.10 e software. Clustering result was based on UPGMA method. The result showed that, 291 polymorphism bands were detected by 40 pairs of SSR markers and the average numbers of polymorphism bands per primer was 7. The PIC value of each SSR loci ranged from 0.53 ~ 0.98 with an average value of 0.76. Genetic similarity coefficient of the 43 rapeseed samples distributed mainly between 0.66 and 0.76. It shows that the tested rapeseed materials are with low genetic diversity and a relatively high genetic similarity. Thus, it's of great importance to enhance the selection of Sclerotinia sclerotiorum resistant lines to broad the genetic background of current ones for further Sclerotinia sclerotiorum resistance breeding.
