摘要
WRKY转录因子是植物界中存在的一类较大的基因家族,参与植物在生物和非生物逆境胁迫中的多种应答。为了研究在低温、高温和盐等非生物胁迫对辣椒CaWRKY13基因表达的影响,以豫椒101为实验材料,在对辣椒CaWRKY13基因克隆的基础上,利用生物软件对其编码蛋白进行分析和构建进化树。结果表明,同源扩增获得的序列含有一个702 bp完整ORF框;对其编码蛋白分析发现,只含有一个WRKY结构域,属于WRKY13家族。亚细胞定位在细胞核中,分子进化树表明与茄属亲缘关系最近,相似性达到92%。荧光定量PCR分析表明,与对照相比,低温、高温和盐等非生物胁迫都能诱导CaWRKY13基因表达,表明CaWRKY13基因在应对低温、高温和盐等非生物胁迫方面具有重要作用。
WRKY transcription factor is a larger gene family that exists in the plant kingdom, which takes part in a variety of plant responses to the biotic and abiotic adversity stress. In order to study the influence on pepper CaWRKY13 gene expression under the abiotic stress, such as low temperature, high temperature and salt stress,and pepper 101 as experimental materials, its encoding protein was analyzed and the evolutionary tree was constructed using software on the basis of CaWRKY13 gene cloning in the hot pepper. The results showed that the homologous expansion of sequence contained a 702 bp complete ORF box. Analysis of its coding protein found that only one WRKY domain structure belonged to the family of WRKY13, named CaWRKY13. Subcellular localization was in the nucleus. The molecular evolutionary tree revealed that it had the closest relative with solanum and the similarity reached 92%. Fluorescence quantitative analysis showed that compared to the control,abiotic stresses such as low temperature, high temperature and salt can induce CaWRKY13 expression, as well as showed that CaWRKY13 gene played an important role in responsing to abiotic stress aspects, such as low temperature, high temperature and salt.
