摘要
为了探讨香菇中疏水蛋白hyd1基因在高温胁迫下的变化情况,进而研究提高香菇对高温的耐受性。本研究首先找出香菇中疏水蛋白hyd1基因序列,筛选出耐高温的香菇菌株18N44和高温敏感型的菌株18为研究对象,比较了高温胁迫下疏水蛋白hyd1基因在香菇两菌株中的相对表达量差异,最后对hyd1基因所编码的氨基酸序列进行了生物信息学分析。实时荧光定量PCR结果显示,在整个高温胁迫期间(4 h除外),香菇菌株18N44的hyd1基因表达量显著高于18;生物信息学分析结果显示,香菇hyd1基因所编码的蛋白质由127个氨基酸组成,属分泌型蛋白,含多个功能位点,与平菇的亲缘关系较近。hyd1基因高温胁迫下表达与生物信息学分析可为香菇耐高温相关机制的进一步研究提供理论参考。
In order to study the relative expression of hyd1 gene during high temperature stress in Lentinula edodes for improving hot-tolerance of it. The sequence of hyd1 gene was obtained from NCBI and its relative expression in mycelia of two Lentinula edodes strains differing in hot tolerance, 18(hot-sensitive) and 18N44(hot-tolerant), being exposed to high temperature stress were analyzed in this study. Real-time PCR result showed that the relative expression of hyd1 gene in 18N44 was higher than that in 18 during high temperature stress(except 4 h). Bioinformatic analysis showed that Hyd1 protein consisted of 127 amino acid residues and was a secreted protein with several functional sites. This protein was similar with the homologous sequence from Pleurotus ostreatus. These results provided theoretical references for further studies of the regulatory mechanism of high temperature tolerance in Lentinula edodes.
