基于trnH-pbsA序列的8种葱属植物系统遗传多样性分析 被引量:4

Genetic Diversity of 8 Allium Based on TrnH-pbsA Sequence

摘要 本研究主要通过对薤白、蒜、大葱、香葱、蒙古韭、阿拉善韭、宽叶韭、黄花葱8种葱属植物trnH-pbsA基因片段进行PCR扩增并测序,使用clustal X1.83、MEGA6.06等软件进行序列分析并构建系统进化树。PCR扩增显示宽叶韭未检出条带,其余7种葱属植物的trnH-pbsA序列长度在618 bp到626 bp之间,经MEGA6.06软件处理分析得到636个位点,序列中共501个保守位点,107个变异位点和104个简约信息位点。GC含量在34.9%~35.8%之间,种间的遗传距离在0.000~0.166之间,同源性在80%~99%之间。分子进化树表明,薤白与其余7种葱属植物分开,蒙古韭和阿拉善韭自展支持率为48%,蒜和香葱自展支持率为18%。薤白与其余7种葱属植物亲缘关系较远,大葱和香葱亲缘关系较近。 We used 8 kinds of Allium species as research materials, including Allium macrostemon Bunge, Allium sativum L., Allium fistulosum, Allium ascalonicum, Allium mongolicum Regel., Allium flavovirens L., Allium hookeri and Allium condensatum L. PCR was performed using TrnH-pbsA primers pairs. TrnH-pbsA sequences were aligned and proofread with Clustal X1.83 and the Lasergene. Neighbor-joining(NJ) trees were generated by MEGA6.06. The results showed that there were 7 Allium were detected, the length of the amplified trn H-pbs A sequences were in a range of 618 and 626 bp. The length of the final alignment was 636 bp. There were 501 invariable sites, 107 variable sites and 104 singleton variable sites. The GC content of trnH-pbsA sequences ranged from 42.0% to 43.2%. The genetic distance distance was in a range between 0.000 and 0.166. The homology of trnH-pbsA ranged from 80% to 99%. The results of Phylogenetic tree showed that the bootstrap of Allium mongolicum Regel. and Allium flavovirens was 48%. The bootstrap of Allium sativum L. and Allium ascalonicum was 18%. According to the result, the relationship between Allium macrostemon Bunge and the other kinds of Allium was farther, and the relationship between Allium fistulosum and Allium ascalonicum was nearer.
