摘要
本研究应用ISSR分析了来自5个不同种源地的73份无患子种质的遗传多样性。ISSR共扩增出条带119条,其中多态性条带105条,多态性百分率为88.24%;表明不同地区的无患子种质间存在较高的多态性,遗传多样性较为丰富,各地区种质间的遗传差异较大;5个无患子种群间的遗传距离变化范围在0.032 4~0.081 0之间;UPGMA聚类分析表明,73份无患子种质可分为2大类,组A由安徽黄山、浙江台州、江西吉安和福建建宁组成;组B仅含江西龙南。分布于浙江台州与分布于江西吉安的无患子种质亲缘关系最近,分布于安徽黄山和江西龙南的无患子种质亲缘关系最远。ISSR方法能有效地阐明不同地区无患子种质的多样性,为更好地利用无患子优异种质资源进行育种提供参考。
This research investigated the genetic diversity of 73 germplasms of Sapindus mukorossi Gaertn from 5areas by ISSR. Using ISSR primer pairs, we amplified 119 DNA bands and 105 ones of them showed polymorphism, which means there were abundant genetic diversity among these S. mukorossi germplasms from different areas; genetic distances among these five sapindus groups ranged from 0.032 4 to 0.081 0; UPGMA cluster analysis showed the presence of two major clusters: cluster A that contained the populations from Anhui Huangshan(AHHS), Zhejiang Taizhou(ZJTZ), Jiangxi Ji'an(JXJA) and Fujian Jianning(FJJN), and cluster B that only contained the cultivars from Jiangxi Longnan(JXLN). The cultivars from ZJTZ and JXJA showed the closest genetic relationship, while the cultivars from AHHS and JXLN showed the farthest genetic relationship. ISSR methods were able to effectively illustrate the genetic diversity among different cultivars of sapindus plants, which provided reference for breeding works by better using the germplasm resources of Sapindus mukorossi.
