摘要
本研究从水芹中克隆得到一个HSP基因Oj HSP90,生物信息学分析表明:水芹Oj HSP90基因全长2 100 bp,编码699个氨基酸;Oj HSP90蛋白的理论分子量为80.3 k D,等电点为5.01,是一种亲水性的稳定蛋白;Oj HSP90蛋白的二级结构中,α-螺旋占52.93%,延伸链占16.74%,β-折叠占5.87%,无规则卷曲占24.46%;该蛋白的N端含有1个HSP90超家族蛋白保守的结构域YSNKEIFLRE,能够与ATP结合;进化树分析表明,Oj HSP90蛋白与拟南芥At HSP90蛋白和烟草Nt HSP90的相似度最高,分别为91.99%和92.13%。本研究旨在为进一步研究HSP90基因的功能以及研究水芹耐高温途径提供理论参考。
In this study, an Oj HSP90 gene was cloned from the cress(Oenanthe javanica) plant. Bioinformation analysis showed that the full length c DNA of Oj HSP90 was 2 100 bp and encoded 699 amino acids with a molecular weight of 80.3 k D and an isoelectric point of 5.01. The protein is hydrophilic and stable. The secondary structure of Oj HSP90 protein comprises of 52.93% alpha helix, 16.74% extending chain, 5.87% beta turn and24.46% random coil. The N-terminal of the protein contains a ATP-binding domain YSNKEIFLRE which is conservative in the HSP90 super-family protein. Phylogenetic tree analysis indicates that Oj HSP90 shares higher similarity with Arabidopsis At HSP90(91.99%) and tobacco(Nicotiana tabacum) Nt HSP90(92.13%). This will be the basis for further investigating the function of the HSP90 gene and the high temperature tolerance pathway of cress(Oenanthe javanica).
