摘要
本研究以粗皮桉无菌苗下胚轴为外植体,通过对不同激素组合的优化,研究不同激素处理对粗皮桉体胚发生的影响,初步建立了愈伤组织诱导及体胚发生最适合的激素浓度与种类。结果表明,0.005 mg/L TDZ和0.1 mg/L NAA激素组合诱导愈伤率最高,外植体愈伤组织诱导率达到97.5%,且该激素组合可以使体胚直接发生。而对体胚诱导效果最好的是1.0 mg/L 6-BA和0.1 mg/L NAA,诱导率为45.0%。本研究为粗皮桉体胚诱导提供借鉴与参考。
In this study, using stems of Eucalyptus pellita grandis as explants to form callus, which treated with different plant biological regulator was optimized with several hormone combinations for inducing stems of Eucalyptus pellita grandis to form somatic embryogenesis and callus embryogeny. The results showed that 0.005 mg/L TDZ and 0.1 mg/L NAA is the best combinations for inducing stems of Eucalyptus pellita grandis to form callus,and 97.5% of explants formed callus, and direct somatic embryogenes can been indicued. The best combinations for somatic embryo induction is 1.0 mg/L 6-BA and 0.1 mg/L NAA, 45.0% of callus formed adventitious bud. With this work, we lay the foundation for the efficient somatic embryogenesis and shoot regeneration in Eucalyptus pellita.
