摘要
由于植物瞬时表达系统具有经济、安全、快速、高效的特殊优势,已经成为药用蛋白表达和生产的最理想的"分子工厂"。我们利用根部真空侵染法在烟草(Nicotiana tabatum L.)中瞬时表达了报告基因GFP,并利用半定量RT-PCR和Western Blot对外源蛋白的表达水平进行了分子水平上的检测。结果表明外源蛋白GFP在烟草(N.tabacum)中成功获得了表达。此外,我们优化了新表达体系的各种侵染条件,为利用新型生物技术生产药用蛋白提供了帮助。
Plant transient expression system has became the best ' molecular factory'' of pharmaceutical proteins expression and production, since the system have these special advantages of economical, safty, fast and high efficiency. Here, we used root vaccum inoculation to express reporter gene GFP in tobacco(Nicotiana tabatum L.)plants by means of transient expression, the expression level of foreign protein was detected by Semi-quantitative RT-PCR and Western Blot as well. The results showed that the foreign protein GFP was successfully expressed in tobacco(N. tabacum) plants. In addition, we optimized various inoculation conditions of the new transient expression system, and provided a help for the production of pharmaceutical proteins using the new biotechnology.
