摘要
本研究采用主基因+多基因混合遗传模型分离分析法对大豆冠层的叶面积指数(leaf area index,LAI)、无截取散射(diffuse none-interceptance,DIFN)、平均叶倾角(mean leaf angle,MTA)3个参数进行遗传分析,并利用Win QTL Cartographer Version 2.5程序的复合区间作图法(CIM)及完备区间作图法QTL Ic IMapping软件进行QTL定位和上位性互作分析。染色体片段置换系(chromosome segment substitution lines,CSSL)群体冠层参数中,LAI最适模型为4MG-AI,DIFN最适模型为4MG-A,MTA两个最适模型为2MG-Duplicate和2MG-Inhibiting,重组自交系(recombinant inbred lines,RIL)群体冠层中LAI最适模型为4MG-EEEA,DIFN最适模型为4MG-A,MTA两个最适模型为2MG-Duplicate和2MG-Inhibiting。两套群体共检测到2个LAI相关的QTL位点,分别位于A1、I连锁群上。检测到22个与DIFN相关的QTL位点,分别位于A1、D1b、F、J、L、N、M、A2、K、B2、I、D1a连锁群上。检测到7个MTA相关的QTL位点,分别位于C2、E、I、A1、D1a连锁群上。CSSL群体冠层参数数据分析中发现,q LAI-A1-1和q DIF,N-A1-1同时在A1连锁群上的3 691~34 163 kb处检测到,q DIFN-A1-2和q MTA-A1-1同时在A1连锁群上的38 669~40 501 kb处检测到,两个位点可能为一因多效位点。仅在RIL群体冠层参数中发现6对QTL间存在着互作效应,在J、A1、B1、F、B2、E、D2连锁群上,在J连锁群上的q DIFN-J-1与A1连锁群上的q DIFN-A1-1和B1连锁群上的q DIFN-B1-1同时发生互作。
The main gene and multiple gene mixed inheritance model method was used to analysis the leaf area index(LAI), diffuse none-interceptance(DIFN) and the mean leaf angle(MTA) of soybean canopy. QTL mapping and epistatic interaction analysis were performed by composite interval mapping(CIM) method of Win QTL Cartographer Version 2.5 and inclusive composite interval mapping(ICIM) method of Ic IMapping Version 3.3 software.In chromosome segment substitution lines(CSSL) population, the best suitable model of LAI is 4MG-AI; the best suitable model of DIFN is 4MG-A; two most suitable models of MTA are 2MG-Duplicate and 2MG-Inhibiting. In recombinant inbred lines(RIL) population, the best suitable model of LAI is 4MG-EEEA; the best suitable model of DIFN is 4MG-A; two most suitable models of MTA are 2MG-Duplicate and 2MG-Inhibiting. Two LAI QTLs were detected on linkage group A1 and I in both populations. Twenty two DIFN QTLs were detected on linkage group A1, D1 b, F, J, L, N, M, A2, K, B2, I and D1 a. Seven MTA QTLs were detected on linkage group C2, E, I, A1 and D1 a. In CSSL population, q LAI-A1-1 and q DIFN-A1-1 were detected at the similar region on linkage group A13 691~34 163 kb, q DIFN-A1-2 and q MTA-A1-1 were also detected at the closed genome region of 38 669~40 501 kb of linkage group A1. These two intervals may be one gene with pleiotropic effects. Six pairs epistatic QTL were detected on linkage group J, A1, B1, F, B2, E, and D2. For q DIFN-J-1 on linkage group J, it showed simultaneous interactions with the QTL on inkage group A1 and inkage group B1 at the same time.
