摘要 磺基转移酶基因是一个基因超家族,它对植物的生长发育和抗病等方面有着重要的作用。为了研究OsCST5 (LOC_Os08g20130)在水稻中的功能,本研究对水稻OsCST5基因进行生物信息学分析并检测其在不同组织中的表达模式。结果表明,水稻OsCST5基因CDS全长1 008 bp,编码产物为一个含335个氨基酸的蛋白质,其分子量为37 320.70。Os CST5蛋白具有典型的磺基转移酶-3结构域,不存在跨膜结构域,也不含信号肽;进一步通过OsCST5共表达基因的GO富集和KEGG富集分析,结果显示OsCST5共表达基因主要参与抗病反应的代谢通路,通过启动子顺式元件分析发现OsCST5启动子区域含有抗病反应和发病相关基因的启动子结合位点。qRT-PCR检测结果表明OsCST5在胚芽中的表达量最高,其次是根。综上所述,OsCST5可能参与了水稻的抗病机制,为进一步研究OsCST5在水稻中的免疫机制提供了理论依据。 The sulfotransferase gene is a gene superfamily that plays an important role in plant growth and development, and resistance to disease. In order to study the function of OsCST5(LOC_Os08 g20130) in rice, this study carried out bioinformatics analysis of rice OsCST gene and examined its expression pattern in different tissues. The results showed that the CDS of rice OsCST5 gene was 1 008 bp in length and encoded 335 amino acids. The molecular weight of the protein was 37 320.70, with a typical sulfotransferase-3 domain, but it did not have transmembrane domain and signal peptide. The results of GO enrichment and KEGG enrichment indicated that the OsCST5 co-expressing gene was involved in the metabolic pathway of disease resistance. Promoter cis-element analysis revealed that the OsCST5 promoter region contained a promoter-binding site for disease resistance and pathogenesis-related genes. PCR results suggested that OsCST5 had the highest expression in germ, followed by root. In summary, Os CST5 might be involved in the disease resistance mechanism of rice, providing a theoretical basis for further study of the immune mechanism of OsCST5 in rice.
