用于杏种质资源遗传多样性分析的SSR引物的筛选 被引量:5

Screening of SSR Markers for Genetic Diversity Analysis of Apricot Germplasm Resources

摘要 由于杏种质资源的分子标记急缺,本研究利用普通桃、樱桃和扁桃植物SSR引物对杏种质进行遗传多样性分析,筛选出可用于杏种质的SSR引物。结果表明,普通桃、樱桃和扁桃植物的82对SSR引物中有56对引物在杏种质可有效扩增,27对引物具有多态性;其中普通桃22对引物具有多态性,樱桃3对引物具有多态性,扁桃2对引物具有多态性。筛选出的27对SSR引物对83份杏种质进行荧光毛细管电泳检测,共检测到203个等位基因(Na),平均每个位点7.52个;多态信息含量(PIC)值为0.67~0.94,均值为0.84,表明所选SSR引物均为高度多态性引物。本研究结果表明近缘植物SSR分子标记在杏种质资源中具有较高的通用性,其中普通桃SSR引物在杏种质多态性最高,这些多态性引物有助于中国杏种质资源的遗传多样性分析。 Due to the shortage of mo lecular markers of apricot germplasm, SSR primers from Prunus persica,Prunus avium and Amygdalus communis were used to analyze the genetic diversity of apricot germplasm in this present paper, and SSR markers suitable for apricot were screened out. The results showed that 56 pairs of SSR primers of the 82 pairs of SSR primers from Prunus persica, Prunus avium and Amygdalus communis could be effectively amplified in apricot, in which 27 pairs were polymorphic, including 22 pairs from Prunus persica, 3 pairs from Prunus avium, and 2 pairs from Amygdalus communis. A total of 203 alleles(Na) were detected in 83 apricot germplasms by fluorescence capillary electrophoresis by 27 pairs of SSR primers, with an average of 7.52 alleles per locus. Polymorphic information content(PIC) value ranged from 0.67 to 0.94, with an average of 0.84,indicating that the selected SSR primers were all highly polymorphic. The results of this study suggested that the SSR molecular marker of closely related plants has high versatility in apricot germplasm resources. Among them,SSR primers of Prunus persica had the highest polymorphism in apricot germplasm. These polymorphic primers were helpful for genetic diversity analysis of apricot germplasm resources in China.
