摘要
通过33对SSR引物对7份双孢蘑菇栽培菌株基因组DNA进行PCR扩增,共检测出62个等位基因,每对引物的等位基因数为2~4个,平均为2.48个。其中25对引物在7个双孢蘑菇基因型中存在明显多态性,占检测引物总数的75.76%。SSR引物仅能将7份菌菇材料区分为4类。其中,国内褐色双孢蘑菇主栽菌株棕秀一号、LY、LC的遗传相似系数为1,与国外褐色双孢蘑菇菌株MG遗传相似系数为0.645 2。国内褐色双孢蘑菇与国外褐色双孢蘑菇菌株MG遗传背景存在差异,国内褐色双孢蘑菇菌株具有相似的遗传背景。结果解释了国内褐色双孢蘑菇菌株遗传背景狭窄,多样性差,选育褐色双孢蘑菇优良栽培菌种具有非常现实的意义。
33 pairs of SSR primers were used to amplify the genomic DNA of 7 kinds of industrial leading cultivated A. bisporus strains, and 62 alleles were detected. The number of alleles in each pair of primers was 2~4, and the average number was 2.48. 25 pairs of primers showed obvious polymorphism in 7 A. bisporus strains genotype,which occupied 75.76% of the detected primers numbers. 7 kinds of A. bisporus strains could be classified into 4 genetic groups by SSR primers. The genetic similarity coefficient was 1 for Zongxiu No.1, LY and LC, and the genetic similarity coefficient was 0.645 2 between Zongxiu No.1 and MG. There were differences in genetic background between domestic and foreign strains of Agaricus bisporus. While domestic strains of Agaricus bisporus had similar genetic background. The results explained the narrow genetic background and poor diversity of the domestic strains of Agaricus bisporus, which was very practical to the selection of the best cultivated strains of Agaricus bisporus.
