不同贮藏温度对萱草花粉活力的影响 被引量:7

Effects of Different Storage Temperatures on Pollen Viability of Hemerocallis fulva

摘要 为解决萱草杂交育种过程中花期不遇、异地杂交等问题,以‘金娃娃’为试材,采用TTC染色法对‘金娃娃’花粉分别在高温干燥、高温湿润两种不同环境条件下1 d内花粉活力变化进行测定,并研究了25℃、4℃、-20℃、-75℃四个贮藏温度条件对萱草花粉生活力的影响情况。结果表明:1 d内早晨7时萱草花粉活力达到最高值,花粉采集应选择高温湿润的环境条件时为佳;不同贮藏条件下,花粉活力变化有很大差异。常温(25℃)贮藏时,花粉活力下降最快,贮藏28 d后全部丧失活力;4℃贮藏条件下保存7 d时活力可达到68.84%,利于短期贮藏;若长期贮藏,则应选择-75℃的温度条件,-75℃贮藏条件下保存42 d后‘金娃娃’花粉活力仍可达到39.96%。本研究为萱草花粉的种质资源保存和杂交育种工作提供了参考依据。 In order to overcome the problem of asynchronous flowering period and cross-breeding in different places in the process of hybrid breeding of Hemerocallis fulva,using’Jin Wawa’pollens as materials,the pollen viability of’Jin Wawa’in one day under two different environmental conditions of high temperature drying and high temperature wetting was tested by means of TTC staining,and effects of storage temperature conditions of25℃,4℃,20℃,and-75℃on Hemerocallis pollen viabilities were researched.The results showed that the pollen viability of Hemerocallis fulva was highest at 7:00 A.M.in a day,and the pollen collection should be preferred when the temperature was high and humidity was wet.The viability of pollens was different under different storage conditions.At room temperature(25℃),pollen vitality decreased fastest,and and all pollen vitality was lost after28 days storage.When stored at 4℃for 7 d,the pollen vitality could reach 68.84%,which was favorable for short-term storage.If stored for a long time,the pollen vitality of’Jinwa’could still reach 39.96%after storage at-75℃for 42 days.This study would provide a reference for gemplasm resources conservation and cross breeding of Hemerocallis fulva.
