混合基因型群体花斑种皮高油酸花生新品系筛选及KASP鉴评 被引量:3

Screening and KASP Evaluation of New High-Oleic Acid Peanut Lines with Varigated Testa in Mixed Genotype Population

摘要 本研究采用高油酸粉皮花生G63品系为母本,普通油酸花斑种皮花生VG-01品系为父本配置杂交组合,借助分子标记筛选到具有表型标记的高油酸花生新品系。通过等位基因特异性多聚酶链反应(allele specific polymerase chain reaction, AS-PCR)检测F4群体基因型分布,结果表明该群体包含Aabb、aaBb和AaBb三种杂合基因型。通过近红外光谱技术(near infrared spectroscopy, NIR)检测F4:5家系的油酸含量,结果表明油酸NIR值小于80%的家系占42.5%,大于等于80%的占57.5%。采用近红外光谱技术在1 375株中检测出产量大于对照‘冀花4号’43.53%,油酸含量介于69.49%~88.05%的单株188株,其中小果型有42个,中果型有129个,大果型有17个,果型大部分为普通型或茧型,部分为斧头形、葫芦形或蜂腰形。竞争性等位基因特异性PCR (kompetitive allele specific PCR, KASP)检测结果表明,188个候选品系中,品系18-A-08、18-A-18、18-B-01、18-B-16和18-B-26基因型全部为aabb。品系比较试验表明,品系18-B-16、18-B-26较对照‘冀花4号’分别提高2.24%和1.37%,在p=0.05和p=0.01水平上均差异不显著。对品系18-B-16和18-B-26的农艺性状考察结果表明,18-B-16和18-B-26的株型分别属于半匍匐型和直立型,油酸含量分别为79.6%和78.53%,亚油酸含量分别为8.57%和11.69%,油酸/亚油酸(O/L)分别为9.28和6.72;在荚果性状方面,18-B-16和18-B-26果嘴差异不明显,果腰差异明显,网纹差异不明显,种皮颜色均为花斑性状。本研究获得2个花斑种皮高油酸花生新品系,该批品系具有较高的营养价值及医疗保健价值,因此本研究对于丰富河北省高油酸花生种质资源以及促进高油酸花生产业化发展具有重要的现实意义。 In this study,the high oleic acid pink testa peanut G63 line was used as the female parent,and the common oleic acid variegated testa peanut VG-01 line was used as the male parent,some novel strain of high oleic acid peanut with phenotypic marker was selected by molecular marker assisted selection.The genotype distribution of F4 population was detected by allele specific polymerase chain reaction(AS-PCR),the results showed that the population contained three heterozygous genotypes of Aabb,aaBb and AaBb.The oleic acid content of F4:5 family was detected by near infrared(NIR).The results showed that the oleic acid NIR value was less than 80%of the family accounted for 42.5%,and the 80%or more accounted for 57.5%.1 357 lines were detected by near-infrared technique,the result demonstrated that the yield of 188 offspring was 43.53%higher that of the control Jihua4,and the oleic acid content was 69.49%~88.05%,of which 42 were small fruit type,129 were medium,and 17 were large.Most of the fruit types are common or cocoon type,and some are axe,gourd or bee waist.Competitive allele specific PCR(KASP)test results showed that among the 188 candidate lines,the genotype of line 18-A-08,18-A-18,18-B-01,18-B-16 and 18-B-26 were all aabb.The line comparison test showed that the line 18-B-16 and18-B-26 were respectively increased by 2.24%and 1.37%compared with that of control,and the difference was not significant at p=0.05 and p=0.01.The results of agronomic traits of line 18-B-16 and 18-B-26 showed that the plant types of 18-B-16 and 18-B-26 belonged to semi-creep and erect,and the oleic acid content was 79.6%and78.53%,the linoleic acid content was 8.57%and 11.69%,and the oleic acid/linoleic acid(O/L)was 9.28 and 6.72,respectively.In terms of pod traits,the difference between 18-B-16 and 18-B-26 fruit mouths was not obvious,the difference in fruit waist was obvious,the difference in texture was not obvious,and the color of seed coat was verigated.In this study,two new lines of high oleic acid peanuts were obtained,which have high nutritional value and health care value.This study has important practical significance for enriching the high oleic peanut germplasm resources and promoting the industrialization of high oleic peanuts in Hebei Province.
