Development and Identification of Rhododendron hybridum EST-SSR Primers based on Transcriptome Sequencing

摘要 杜鹃(Rhododendron hybridum Hort.)是一种重要的观赏和药用植物,基于转录组测序的EST-SSR引物开发,对其遗传多样性分析和野生种质资源保护及开发利用具有重要意义。本研究基于高通量转录组测序技术,以杜鹃花根、茎、叶和花混合样本为材料进行测试分析,筛选出21对多态性SSR引物,并对其在1个野生杜鹃花种群中的24个样本进行了多样性检测,所有位点均显示出了高度的多态性。每个位点的等位基因数从2个变化到13个,观测杂合度变化范围为0.029~1.000,期望杂合度为0.028~1.000。这些EST-SSR标记的开发为杜鹃花遗传多样性分析、遗传图谱构建、种质资源开发利用及分子辅助育种提供宝贵资源。 Rhodo dendron hybridum Hort.is an important ornamental and medicinal plant.The development of EST-SSR primers based on transcriptome sequencing is of great significance for its genetic diversity analysis and protection and utilization prospects of wild Rhododendron hybridum resources.Based on high-throughput transcriptome sequencing technology,this study takes mixed sample of rhododendron roots,stems,leaves and flowers as a material for test and analysis to screen 21 pairs of polymorphic SSR primers and test the diversity of 24 individuals from a wild Rhododendron population.All sites show a high degree of polymorphism.The number of alleles locus at each site is varied from two to thirteen.The observed heterozygosity is ranged from 0.029 to 1.000,and the expected heterozygosity is ranged from 0.028 to 1.000.The development of these EST-SSR markers provides valuable resources for genetic diversity analysis,genetic map construction,development and utilization of germplasm resources,and molecular-assisted breeding of rhododendrons species.
