摘要
芬芳安息香(Styrax odoratissimus)为安息香科(Styracaceae)安息香属(Styrax)落叶小乔木,在材用、药用、园林观赏中都存在一定的价值。为了给芬芳安息香的大规模育苗提供技术支持,并对安息香属其它植物的快速繁育提供参考,本研究以芬芳安息香种子为实验材料,探索较适合其生长的组培条件。结果表明,芬芳安息香较适宜芽增殖的培养基为MS+1 mg/L 6-BA (6-苄基腺嘌呤)+1 mg/L IAA (吲哚乙酸)+0.3 mg/L KT (激动素),增殖系数为6.12,叶色绿且生长旺盛,茎秆粗壮,整体生长状态良好。根诱导同时采用了一阶段诱导和二阶段诱导两种方式,其中一阶段诱导中,较适宜生根的处理为1/2MS+3 mg/L IAA,生根率为30%;二阶段诱导较适宜生根的处理为MS+300 mg/L IAA处理3 d,后转移至1/2MS培养基中,根系生长健壮,且二阶段诱导的生根率远高于一阶段诱导,为72.5%。试管苗移栽至配制比例为1∶1∶1的泥炭、蛭石、珍珠岩混合基质中,成活率为40%。本实验对芬芳安息香无性快繁和产业化育苗有一定的借鉴意义。
Styrax odoratissimus is a deciduous and resource-oriented aromatic plant with ornamental,material and medicinal value.In order to provide technical support for the large-scale breeding of Styrax odoratissimus,and to provide guidelines for tissue culture of other plants in%Styrax,the seeds of Styrax odoratissimus were used as tissue culture materials to explore the most suitable conditions for its growth.The results showed that MS medium supplemented with 1 mg/L 6-BA,1 mg/L IAA and 0.3 mg/L KT was the optimal medium for bud multiplication with the proliferation coefficient reaching 6.12.Two methods were applied for rooting treatment:one step induction and two steps induction.In one step induction,the most suitable treatment was the 1/2 MS supplemented with 3 mg/L IAA,resulting in the rooting rate approaching 30%.While,in two steps induction,the most suitable treatment for rooting was the MS medium supplemented with 300 mg/L IAA for 3 days,and then transferred into1/2 MS,generating long and thick roots with the rooting rate up to 72.5%,which was remarkably higher than one step induction.When transferred to a mixture of peat,vermiculite and perlite with a ratio at 1∶1∶1,the survival rate of rooting plantlets was 40%.Our research provides guidelines for the asexual propagation and industrialized production of Styrax odoratissimus.
