摘要
通过比较卡介苗 (BCG)、卡介苗多糖核酸提取物 (BCG_PSN)以及热休克蛋白65(HSP65)对脐血单核细胞转化的树突状细胞成熟的影响 ,以探讨BCG及其有效成分对新生儿免疫功能的作用。经体外分离 ,诱导19例健康足月新生儿CD14 +单核细胞分化成为未成熟树突状细胞 ,并观察BCG、BCG_PSN和HSP65对其成熟的影响。结果 :BCG可通过上调CD83、CD86和MHCⅡ类抗原 ,下调甘露聚糖受体 (MR)促进脐血单核细胞转化的树突状细胞成熟 ,BCG_PSN和HSP65不能促进脐血树突状细胞分化成熟。提示BCG可能通过促进脐血树突状细胞的成熟 ,影响新生儿T细胞功能发育 。
In this study,the comparison of clinical effects was carried out about mycobacterium bovine bacillus Calmette-Guerin (BCG)and its components,including polysaccaride nucleic acid(BCG-PSN)and heat shock protein 65(HSP65),on the maturation of cord blood monocyte-derived dendritic cells(DCs). Because they can help us to understand the BCG and its components effects on the neonatal immunefunction.Purified cord blood CD14+ monocytes isolated by immunomagnetic beads were cultured in serum and hormone-free medium(DMEM-F12)with IL-4 and GM-CSF for 7 days.On the 5th day of culture,BCG and its components were added for further 2 days culture.The phenotype of the cord blood monocyte-derived DCs expressed was assayed with three-color flow cytometry.In comparison with the immature cord blood monocyte-derived DCs,the BCG induced the maturation of cord blood monocyte-derived DCs can significantly up-regulate the expression of CD83,CD86 and MHC class Ⅱmolecules and down-regulate the expression of mannose receptor(MR).But,its components had no effect on cord blood monocyte-derived DCs maturation.It is possible that through promoting DC maturation,BCG can modulate the development of neonatal naive T cell.But its components have no role in the maturation process of cord blood DCs.
