菜粉蝶颗粒体病毒和浓核病毒混合感染菜青虫细胞系

THE MIX-INFECTION OF GRANULOSIS VIRUS AND DENSONUCLEOSIS VIRUS IN PIERIS RAPAE CELL

菜粉蝶颗粒体病毒(Pieris rapae granulosis virus,PrGV)属于杆状病毒科(Baculoviridae)本干状病毒属B亚群。菜粉蝶浓核病毒(Pieris rapae densonucleosis virus,PrDNV)属于细小病毒科(Parvoviridae)浓核病毒属。这两种病毒经常混合感染菜青虫,对菜青虫有很高致死率。我们用PrGV感染幼虫并收集其血淋巴,用以攻毒菜粉蝶细胞(来自虫卵)时,出现PrGV和PrDNV同时感染了菜粉蝶细胞的现象。 给菜青虫2龄幼虫喂饲涂有PrGV的苞菜叶,第3天收集病虫取其血淋巴。

In this experiment, we employed the infected larvae hemolymph as inoculum, challeng to Pieris rapae cell ( Pr cell) . First, Pr cells showed obvious cytopathic effects. They linked each other and clustered. Laterly, the nuclei of infected cells underwent swelled, the cytoplasm membrance became vague and the granular increased in cytoplasm under light microscope.Electron microscope showed a number of PrGV capsules, virions and inclusions being occluded in the cytoplasm of Pr cell. Longitudinal section of mature capsules are oval with a size of ca 400×250nm,containing a virion which is ca 250×50nm in centre. Some abnormal capsules also have been found in Pr cell, their size were different from normal capsules.Simultaneously, part cells were infected by Pr densonucleosis virus. The infected cells showed the typical densonucleosis virus symptom, the nuclei of infected cells swelled and filled with high electron dense materials. The nuclear membrance began to disintegrate and a larger number of dispersed round bodies were released from nucleus. It was worthy that there was a nucleus showing typical densonucleosis virus symptom and PrGV capsules in cytoplasm of the same cell.
