抗变形链球菌鸡卵黄抗体抑制变形链球菌产酸能力研究

摘要 目的通过对抗变形链球菌Ⅰ、Ⅲ型鸡卵黄抗体(IgY)影响变形链球菌产酸能力的实验研究,探索抗变形链球菌鸡卵黄抗体防龋的机制。方法用含不同浓度抗变形链球菌鸡卵黄抗体TS培养基厌氧培养变形链球菌S.mutans MT6R(血清C型、遗传Ⅰ型)、远缘链球菌S.Sobrinus 6715(血清g型,遗传Ⅲ型)及其混合菌株,用数值式酸度计检测培养基上清液的pH值。数据作方差分析。结果抗变形链球菌Ⅰ、Ⅲ型鸡卵黄抗体能抑制各实验组细菌培养基pH值的下降,经方差分析,实验组pH值随着特异抗体浓度的增加而上升,其最佳抑制浓度为1:2,此时与对照组比较有非常显著的差异;当抗体浓度为1:32时,其pH值与对照组无显著差异(P>0.05)。结论抗变形链球菌鸡卵黄抗体可以有效抑制变形链球菌利用糖产酸的能力。 Objective To evaluate the inhibitory effect of immunoglubulin yolk (IgY) against Streptococcus Mutans on acid production of Mutans Streptococci, and determine the probably anticatries mechanism of IgY.Methods Cells of S.mutanMT6R, S.sobrinus6715 and S.mutansMT6R together with S.sobrinus6715 were cultivated on TS culture medium supplemented with IgY of different concentration.The tubes were incubated at 37℃ under microaerophilic condition (90% N2, 10% CO2) for 30 hours. The value of pH of culture medium substrate were measured with a digital acidimeter. The data were analzed by an analysis of variance (AMOVA) . Results IgY against Streptococcus Mutans Ⅰ 、 Ⅲ can prevent the pH of experimental group from droping.The pH of experimental group rosed with the antibodies concentration increased. When the IgY' s titer is 1:2, the pH of experimental groups are different from the control group' s significantly (P< 0.01) .when the titer is 1 :32, there' s no significiantly difference between the experimental and control group (P> 0.05) .Conclusion The results suggest that the specific immunoglobulin yolk against Mutans Streptococci (IgY) can inhibit Mutans Streptococci' s of acid produce ability effectively, and the best concentration is 1:2 of the original.
