db-cAMP对胚胎干细胞来源的神经细胞酪氨酸羟化酶mRNA水平的影响

摘要 目的 体外诱导胚胎干细胞 (ES细胞 )定向分化为多巴胺能神经元并促进其长期存活和特性维持。方法 以全反式维甲酸 (ATRA)诱导 ES细胞定向分化为神经细胞 ,培养液中加入双丁酰基环腺苷磷酸 (db-c AMP)继续培养 ,RT- PCR方法检测酪氨酸羟化酶基因 (TH基因 )和 survivin基因的 m RNA水平变化。结果  db-c AMP可上调 ES细胞来源的神经细胞中 TH基因的 m RNA水平 ,在实验早期还可上调 survivin基因的 m RNA水平。结论  db- c AMP可以促进 ES细胞来源的多巴胺能神经元的存活和特性维持 ,抗凋亡可能是其作用机制之一。 Objective To induce the embryonic stem cells(ES cells) to be dopaminergic neurons and to promote the survival and maintainence of these neurons. Methods ES cells were induced by all trans retinoic acid(ATRA) to be neurons. These ES cell derived neurons were cultivated with db cAMP and the mRNA level of Tyrosine Hydroxylase(TH) and survivin were detected by RT PCR. Results db cAMP could up regulate the TH and survivin mRNA levels. Conclusion db cAMP can promote the survival or maintainence of ES cell derived dopaminergic neurons. Anti apoptosis is the probable mechanism.
