血小板第4因子对同基因骨髓移植小鼠造血重建的影响

Effects of Platelet Factor 4 on Hematopoietic Reconstruction of Syngenic Bone Marrow Transplantation Mice

摘要 为探讨血小板第 4因子 (PF4 )对同基因骨髓移植 (BMT)小鼠造血重建的影响及其可能机制 ,分别于照射前 2 0、 2 6 h给小鼠腹腔注射 PF4 2 0 μg/ kg共 2次 ,然后建立同基因骨髓移植小鼠模型 ,分别于 BMT后 +7、 +14 d计数外周血细胞、骨髓单个核细胞 (BMNC)以及脾集落形成单位 (CFU - S) ,检测趋化因子受体 CXCR4的表达。 PF4处理组小鼠 +7、+14 d BMNC以及 +7d CFU- S计数显著高于 BMT组小鼠 (P<0 .0 1或 P<0 .0 5 ) ;BMT后 +14 d,PF4组小鼠 CXCR4的表达显著高于 BMT组 (P<0 .0 5 )。表明 PF4可以加速 BMT后的造血重建过程 ,促进 To explore the effects of platelet factor 4 (PF4) on hematopoietic reconstruction and its mechanism in syngenic bone marrow transplantation (BMT), the syngenic BMT mice model was established. 20 and 26 h before irradiation,the BALB/c mice were subjected to the injection of 20 μg/kg PF4 or PBS twice into abdominal cavity,then they were transplanted with the donor bone marrow nuclear cells (BMNC). On the 7 and 14 day after BMT,peripheral blood cells, BMNC and spleen clone forming unit (CFU S) were counted and the CXCR4 expression was detected. In PF4 treated group,the peripheral blood cell counts, BMNC counts on the +7, +14 day after BMT and the CFU S counts on the +7 day after BMT were higher than in the BMT group ( P<0 01 or P <0 05). The CXCR4 expression levels in PF4 treated groups were higher than BMT group on the 14 day after BMT (P<0 01 or P<0 05) . These suggested that PF4 could accelerate hematopoietic reconstruction of allogenic bone marrow transplantation. One of the mechanisms might be the enhanced CXCR4 expression on BMNC.
