摘要
采用结扎左冠状动脉前降支 30 min,再灌注 4 0 min复制在体大鼠心肌缺血再灌注损伤 (IRI)的模型 ,结扎前 10 min分别舌下静脉注射倒卵叶五加总皂甙 (SAOH) 5 0 m g/kg、 10 0 mg/kg。结果发现 :SAOH能显著降低再灌注心律失常的发生率 ,缩短心律失常持续时间 ;减缓再灌注 30 min时左心功能低下 ,对再灌注后左室压最大上升速率(+dp/dt.max)的改善尤为明显 ;并能显著减少血浆中心肌酶的含量 ,防止心肌酶从胞内漏出 ,提高 SOD活性 ,减少MDA的生成量 ;减轻 IRI时超微结构的改变。由此认为 :SAOH能抑制 IRI过程中心肌电活动紊乱 ,对心肌有明显的保护作用 ,其机制之一可能与其提高抗氧化酶活性 ,减少脂质过氧化反应 。
Myocardial ischemia reperfusion injury (IRI) model was induced by left descending anterior coronary artery (LAD) ligation for 30 min, followed by 40 min of reperfusion. Ten min before LAD ligation Saponin of Acanthopanax Obovatus Hoo (SAOH) 50 mg/kg or 100 mg/kg was administered by the route of sublingual vein injection. The results showed: SAOH could significantly decrease the incidence and duration of ventricular arrhythmia and decrease ventricular disfunction, especially in left ventricular derivative developed force (+dp/dt.max) after 30 min of reperfusion; SAOH could markedly decrease the levels of serum myocardial enzymes, prevent myocardial leakage, increase the ability of superoxide dismutase and reduce contents of the malondialdehyde. SAOH could obviously alleviate the ultrastructural changes in IRI. These findings suggested that SAOH could obviously protect myocardia during IRI, which might be associated with enchancing the ability of anti oxygen free radical and scavenging free oxygen radical.
