近交系小鼠耳蜗钙调素依赖蛋白激酶Ⅱ活性变化

摘要 目的 通过对 C5 7和 BAL B/ c近交系小鼠不同日龄时耳蜗基底膜钙调素依赖蛋白激酶 活性 (calm odulin-dependent protein kinase ,Ca M- PK )的测定 ,探讨其随年龄变化的趋势 .方法 采用 Ca M- PK 活性检测试剂盒以及同位素掺入技术分别于 2 0 ,30 ,6 0 ,90 ,12 0 ,15 0和 180日龄(n=6 )时检测酶活性 ,实验数据采用 SPSS 10 .0统计软件处理 .结果  C5 7和 BAL B/ c近交系小鼠随年龄增大均出现Ca M- PK 活性下降 ,与 2 0日龄相比 ,90日龄时 Ca M- PK 活性显著降低 (C5 7:5 1.3± 0 .3vs 5 2 .5± 0 .5 pkat· g- 1 ,P<0 .0 1;BAL B/ c:5 0 .9± 0 .5 vs5 2 .5± 0 .4 pkat· g- 1 ,P<0 .0 1) ,随着年龄增大 ,Ca M- PK 活性进一步下降 .结论 Ca M- PK 可能参与 C5 7和 BAL B/ AIM To observe dynamic changes of Ca 2+ /cal modulin dependent protein kinaseⅡ (CaM PKⅡ) activity levels in cochlear basilar membrane of C57 and BALB/c inbred mouse strains with age. METHODS CaM PKⅡ activities were measured with CaM PKⅡKit and 32 P incorporation technique of the mice aged 20, 30, 60, 90, 120, 150, 180 days, respectively. RESULTS CaM PKⅡ activity decreased gradually in cochlear basilar membrane of C57 and BALB/c with age. There were significant differences in the 90, 120, 150, 180 day groups compared with the 20 day group, respectively. CONCLUSION CaM PKⅡ may be involved in the signal transduction of age related hearing loss in C57 and BALB/c inbred mouse strains.
