摘要 目的 :评价细菌基因组结构作为种系标志在细菌亲缘关系识别中的应用。方法 :本研究选用 3个属的细菌 :Klebsiel la ,Neisseria以及Vibrio。为得到完整的基因组DNA ,整个提取的操作过程都在琼脂糖凝胶中进行。经proteinaseK除蛋白后 ,基因组DNA置于TE缓冲液中待用。内切酶Ⅰ CeuⅠ是本研究中主要的工具酶。酶切后的基因组DNA经脉冲场电泳分离。结果 :Klebsiella的所有菌株 (两个种 ,即Klebsiellapneumoniae和Klebsiellaoxytoca)都有一个共同的基因组结构 ,而这一“Klebsiella基因组结构”与Escherichiacoli或Salmonella的基因组结构很相似 ,其主要不同之处有 3点 :(1)有 8个rrn操纵子 (Escherichiacoli和Salmonella各有 7个 ) ;(2 )基因组较大 ;(3)Ⅰ CeuⅠE片段易位。以往学术界认为Neisseria具有高度易变和多态的基因组结构。而我们的研究结果表明 ,Neisseria的基本基因组结构是相对稳定的。我们比较研究了 30 0株N .meningitidis,结果仅仅发现了少数几个不同的基因组结构。其中Z2 4 91株代表 5 0 %的群体 (Ⅰ型基因组结构 ) ,MC5 8株代表 2 0 %的群体 (Ⅱ型基因组结构 ) ,N0 4 5株代表 17%的群体 (Ⅲ型基因组结构 )。已知Z2 4 91和MC5 8代表不同的种系群 。
