基于GEO数据库分析AKAP12在胃癌组织中的表达及临床意义

Analysis of expression and clinical significance of AKAP12 in gastric cancer based on GEO database

目的探讨胃癌组织中支架蛋白A激酶锚蛋白12(AKAP12)的表达及临床意义。方法收集2013年1月至2016年12月88例胃癌组织和79例癌旁组织,采用实时定量PCR(QPCR)检测以上组织中AKAP12的表达水平,同时下载NCBI胃癌公共数据集GSE62254中胃癌组织AKAP12表达数据和临床病理资料,分析AKAP12表达与胃癌临床病理特征的关系;根据随访数据分析AKAP12表达与预后的关系,进一步采用Kaplan-Meier Plotter在线生存分析数据库对胃癌组织AKAP12表达与预后的关系进行大数据分析。结果QPCR检测结果显示,胃癌组织中AKAP12的水平为2.880±1.335,高于癌旁组织的1.118±0.508(P<0.05);公共数据集GSE62254中胃癌组织的AKAP12水平为2.149±0.464。AKAP12水平与年龄、N分期和M分期无关(P>0.05),而与Lauren分型、T分期和TNM分期有关(P<0.05)。生存分析显示,AKAP12高表达者的中位无病生存期(DFS)和总生存期(OS)分别为26.0个月(95%CI:24.818~27.182个月)和40.0个月(95%CI:33.077~46.923个月),均低于低表达者的44.0个月(95%CI:38.565~49.435个月)和50.0个月(95%CI:46.472~53.528个月),差异有统计学意义(P<0.05)。在数据集GSE62254中,AKAP12低表达组的DFS和OS均优于高表达组(HR=2.27,95%CI:1.57~3.28,P<0.05;HR=2.29,95%CI:1.57~3.35,P<0.05);进一步采用Kaplan-Meier Plotter在线分析,发现AKAP12低表达组的DFS和OS均优于高表达组(HR=2.18,95%CI:1.71~2.80,P<0.05;HR=2.04,95%CI:1.64~2.55,P<0.05)。结论AKAP12在胃癌组织中高表达,与胃癌的Lauren分型、T分期和TNM分期及预后有关,高表达者的预后较差,有作为胃癌预后预测和病情评估的潜能。

Objective To investigate the expression and clinical significance of the scaffold protein A-kinase anchor protein 12(AKAP12)gene in gastric cancer tissues.Methods Eighty-eight cases of gastric cancer and 79 cases of paracancerous tissue were collected from January 2013 to December 2016.Real-time quantitative PCR(QPCR)was used to detect the expression of AKAP12 in gastric cancer tissues.The expression data and clinicopathological parameters of AKAP12 in gastric cancer were downloaded from the NCBI data set GSE62254.The relationship between AKAP12 expression and clinicopathological characteristics of gastric cancer was analyzed.The relationship between AKAP12 expression and prognosis was analyzed according to follow-up data.The Kaplan-Meier Plotter online survival analysis database was used to analyze the relationship between AKAP12 expression and prognosis of patients with gastric cancer.Results The results of QPCR showed that the AKAP12 level in gastric cancer tissues was 2.880±1.335,higher than 1.118±0.508 in adjacent tissues(P<0.05),and 2.149±0.464 in data set GSE62254.AKAP12 level in gastric cancer tissues was not related to age,N stage and M stage(P>0.05),but related to Lauren classification,T stage and TNM stage(P<0.05).Survival analysis showed that the median disease-free survival(DFS)and overall survival(OS)of 44 patients with high expression of AKAP12 were 26.0 months(95%CI:24.818-27.182 months)and 40.0 months(95%CI:33.077-46.923 months),lower than 44.0 months(95%CI:38.565-49.435 months)and 50.0 months(95%CI:46.472-53.528 months)of 44 patients with low expression of AKAP12(P<0.05).In data set GSE62254,the DFS and OS of patients with low expression of AKAP12 were better than those of patients with high expression(HR=2.27,95%CI:1.57-3.28,P<0.05;HR=2.29,95%CI:1.57-3.35,P<0.05).Further online Kaplan-Meier Plotter analysis showed that the DFS and OS of patients with low expression of AKAP12 were better than patients with high expression(HR=2.18,95%CI:1.71-2.80,P<0.05;HR=2.04,95%CI:1.64-2.55,P<0.05).Conclusion The high expression of AKAP12 in gastric cancer is related to Lauren classification,T stage,TNM stage and prognosis of gastric cancer.The high expression of AKAP12 has a poor prognosis and potential for prognosis prediction and disease evaluation of gastric cancer.
