重组人甲状旁腺激素1-34对豚鼠骨关节炎的改善作用

Effects of recombinant human parathyroid hormone 1-34 on osteoarthritis in guinea pigs

目的:评估重组人甲状旁腺激素1-34(PTH1-34)对豚鼠自发性骨关节炎的作用。方法:采用6月或10月龄豚鼠,每天皮下注射生理盐水或PTH1-34或复泰奥3个月,考察II型胶原C端肽CTX-2、骨钙素OC、组织病理学OARSI评分、骨体积分数BV/TV及骨小梁模式因子Tb.pf等指标。结果:6月龄豚鼠给药后,PTH1-34或复泰奥可降低CTX-2水平、OARSI评分、Tb.pf,提高BV/TV,对豚鼠骨关节炎有改善作用;PTH1-34 30及60 mg/kg改善明显,停药1个月后30 mg/kg组仍可见明显骨改善。10月龄豚鼠PTH1-34给药后雌性动物Tb.pf明显低于对照,认为存在骨改善。结论:PTH1-34对6月龄豚鼠骨关节炎有明显改善作用,对10月龄豚鼠也有一定程度骨质量改善。

Objective:To evaluate the effects of recombinant human parathyroid hormone 1-34(PTH1-34)on spontaneous osteoarthritis in guinea pigs.Methods:Six or ten months-old guinea pigs were subcutaneously injected daily with saline,PTH1-34 or Forsteo for 3 months.The changes of C-telopeptides of type II collagen(CTX-2),osteocalcin(OC),histopathological OARSI score,bone volume fraction BV/TV,and bone trabecular pattern factor Tb.pf were observed.Results:CTX-2 levels,OARSI score and Tb.pf were decreased while BV/TV was increased in 6-month-old guinea pigs after administration of PTH1-34 or Forsteo compared with saline control,which indicated that PTH1-34 or Forsteo could improve guinea pig osteoarthritis and furthermore,the improvement was more significant in guinea pigs with PTH1-34 dosage 30 or 60 mg/kg.Bone improvement was still visible in guinea pigs with PTH1-34 30 mg/kg after 1 month recovery.The Tb.pf level in female 10 months-old guinea pigs was significantly lower than that of the control after PTH1-34 was administered,which was considered that there existed bone improvement.Conclusion:PTH1-34 has a significant improvement effect on spontaneous osteoarthritis in 6 months old guinea pigs,while only a certain degree of improvement in 10 months old guinea pigs.
