摘要
提取分离绿绒蒿属植物高质量的RNA是基因表达、调控与基因工程等研究的基础。通过采用研钵手动研磨的研磨方式处理叶片,采用Trizol法,OMAG试剂盒法,国产天根试剂盒法三种提取方式从绿绒蒿属植物叶片中提取RNA,将不同提取方法提取的RNA通过Nanodrop2000超微量分光光度计和琼脂糖凝胶电泳法进行测定,对RNA的浓度和质量进行分析。结果表明,几种不同提取方法均能获得较为完整的RNA。均可直接用于下游PCR反应,OMAG法提取的RNA纯度和得率均较高;天根试剂盒法提取的RNA纯度不高;Trizol提取的RNA纯度和质量均比较好,但成本较高,本研究对绿绒蒿属植物的分子生物学方面提供了科学依据。
Extraction and isolation of high quality RNA from Artemisia is the basis of gene expression,regulation and genetic engineering.The leaves of Artemisia annua were treated by hand grinding with abrasive bowl.RNA was extracted from leaves of Artemisia annua by three methods that were Trizol method,OMAG Kit Method and Tiangen kit method.RNA extracted by different methods was determined by Nanodrop 2000 ultramicro-spectrophotometer and agarose gel electrophoresis.The concentration and quality of RNA were analyzed.The results showed that a relatively complete RNA could be obtained by several different extraction methods.These methods all could directly used for downstream PCR reactions.The purity and yield of RNA extracted by OMAG method were higher than those by Tiangen kit method,and the purity and quality of RNA extracted by Trizol method were better,but the cost was higher.This study might provide a scientific basis on the molecular biology for Meconpsis Vig.
