海葵附生油脂降解菌的分离鉴定及生长条件优化 被引量:1

Isolation and Identification of An Oil-Degrading Bacterium from Anthopteura midori Uchida et Muramatsu and Optimization in Growth Conditions

摘要 采用油脂中性红平板筛选法,从太平洋绿侧花海葵体腔中分离筛选到一株具有油脂降解能力的菌株XZL-13,依据菌株的形态特征、生理生化特性及分子生物学特征对其归属进行了鉴定,并采用分光光度法探究了该菌株的最适生长条件,研究了该菌株在最适生长条件下的生长曲线及以不同油脂作为唯一碳源的生长情况。结果表明,筛选获得的油脂降解菌XZL-13为蒙氏假单胞菌(Pseudomonas monteilii);最适生长条件为:初始pH值7.5、初始盐度2.0 g·L^-1、培养温度25℃,在此条件下培养7 h,菌落总数达到最大;油脂降解菌XZL-13在以玉米油、花生油或芝麻油为唯一碳源的培养基中生长情况较好。 We isolated and screened an oil-degrading bacterium XZL-13 from Anthopteura midori Uchida et Muramatsu by oil neutral red plate screening method.Moreover,we identified the strain by the morphological characteristics,physiological and biochemical characteristics,and molecular biological characteristics,studied the optimum growth conditions by spectrophotometry,and investigated the growth curve of the strain under the optimum growth conditions and its growth by using different oils as the sole carbon source.The results show that the oil-degrading bacterium XZL-13 belongs to Pseudomonas monteilii.The optimum growth conditions are determined as follows:the initial pH value of 7.5,the initial salinity of 2.0 g·L^-1,and the culture temperature of 25℃.Under above conditions,the maximum colony number can be obtained after cultured for 7 h.The oil-degrading bacterium XZL-13 grows well in the medium by using corn oil,peanut oil,or sesame oil as the sole carbon source.
