摘要
采用荧光光谱法检测甘草酸二钾对核因子κB受体活化因子配体(RANKL)胞外结构域的荧光猝灭作用,并构建人外周血单核巨噬细胞THP-1向破骨细胞样细胞分化模型,探究甘草酸二钾对破骨细胞生成的影响。结果表明,甘草酸二钾对RANKL胞外结构域的内源荧光有明显的猝灭作用,ANS荧光先减弱后逐渐增强,推测甘草酸二钾可能导致RANKL胞外结构域的微结构发生了改变;甘草酸二钾可以抑制破骨细胞样细胞的分化。可以推测,甘草酸二钾与RANKL的相互作用干扰了RANKL与核因子κB受体活化因子(RANK)的结合,从而抑制巨噬细胞向破骨细胞样细胞的分化。
We studied the fluorescence quenching effect of dipotassium glycyrrhizinate on receptor activator of nuclear factor-κB ligand(RANKL)extracellular domain by fluorescence spectrometry,and constructed a differentiation model of human peripheral blood mononuclear cell THP-1 into osteoclast-like cell in order to investigate the effect of dipotassium glycyrrhizinate on the formation of osteoclasts.The results show that dipotassium glycyrrhizinate strongly quenches the endogenous fluorescence of RANKL extracellular domain,and the fluorescence of ANS decreases at first and then gradually enhances.It is speculated that dipotassium glycyrrhizinate may lead to the change of the microstructure of RANKL extracellular domain.Moreover,dipotassium glycyrrhizinate can inhibit the differentiation of osteoclast-like cell.Through above results,we can speculate that the interaction between dipotassium glycyrrhizinate and RANKL interferes with the binding of RANKL and receptor activator of nuclear factor-κB(RANK),thus inhibiting the differentiation of macrophages into osteoclast-like cell.
