花生网斑病菌粗毒素的提取分离及其对寄主 SOD活性、MDA及O2-含量的影响

摘要 利用常规方法提取花生网斑病菌粗毒素,对无菌水、甲醇、无水乙醇、乙酸乙酯4种溶剂及其不同浓度进行筛选,筛选出20%甲醇是溶解花生网斑病菌粗毒素的最佳溶剂;并测定花生网斑病菌粗毒素在接种4种不同抗性的健康花生叶片后0~24 h内叶片的SOD活性、MDA含量和氧离子含量的变化规律。结果表明,接种粗毒素后SOD活性则呈先下降后上升的趋势,且抗性品种的SOD活性始终低于感病品种;抗性品种的MDA含量整体低于感病品种,表明抗性品种的细胞在感染后有较少的过氧化作用,即细胞损伤较少,且抗过氧化能力更强;氧离子含量无明显规律,由此可以推测,氧离子含量的变化受菌丝生长的影响较大。 This study used conventional methods to extract crude toxin of peanut net pathogen,screened 4 solvents including sterile water,methanol,anhydrous ethanol and ethyl acetate with different concentrations,and screened that 20%methanol was the best solvent to dissolve crude toxin of peanut net pathogen.The changes of SOD activity,MDA content and oxygen anion content in peanut leaves inoculated with crude toxin of peanut net pathogen were determined.The results showed that SOD activity decreased first and then increased after inoculation with crude toxin,and SOD activity of resistant varieties was always lower than that of susceptible varieties.MDA content of resistant cultivar was lower than that of susceptible cultivar on the whole,indicating that the cells of resistant cultivar had less peroxidation effect after infection,that is,less cell damage and stronger peroxidation resistance.The oxygen anion content has no obvious rule,so it can be inferred that the ch ange of oxygen anion content is greatly affected by mycelium growth.
