逼尿肌过度活动中RyR2开放程度变化及其发生机制的研究 被引量:1

摘要 目的探讨逼尿肌过度活动(DO)中Ryanodine受体亚型2(RyR2)开放程度的变化及其发生的机制。方法构建膀胱逼尿肌过度活动的动物模型,通过激光扫描共聚焦显微镜检测正常对照组和DO组钙火花,通过Western blot检测正常对照组和DO组大鼠磷酸化RyR2(p-RyR2)和FKBP12.6的表达情况。结果相比于正常对照组,DO组大鼠膀胱逼尿肌细胞钙活动明显降低,FKBP12.6蛋白表达明显增加,而p-RyR2表达明显下降,差异具有统计学意义(P<0.05)。结论DO组FKBP12.6明显增加,p-RyR2表达明显下降,可能是DO逼尿肌RYR2开放降低发生的关键原因。 Objective To explore the variation of ryanodine receptor subtype 2(ryr2)opening and its mechanism in detrusor overactivity(DO).Methods An animal model of overactive detrusor muscle of the bladder was constructed.The spontaneous calcium activity in the normal control group and DO group was detected by laser scanning confocal microscope,and the expression of phosphorylated RyR2(p-RyR2)and FKBP12.6 in the two groups were detected by Western blot.Results Compared with the normal control group,the DO group showed significantly decreased spontaneous calcium activity,increased expression of FKBP12.6 protein,and decreased expression of p-RyR2.Conclusion In the DO group,the expression of FKBP12.6 protein was significantly increased,and the expression of p-RyR2 was significantly decreased,which may be the key reason for the reduction of the RyR2 opening in the detrusor overactivity.
