摘要
本研究将采集自河南某猪场的疑似感染PEDV的猪肠道及其内容物处理后接种于Vero细胞并进行连续体外传代培养。经过S基因遗传进化分析、RT-PCR和间接免疫荧光试验等成功在体外分离鉴定了该毒株,并命名为PEDV HNLB。该毒株能够在Vero细胞上稳定传代并产生典型的细胞病变效应(CPEs),其第6代的病毒滴度达到104.3TCID50/mL。HNLB毒株的成功分离为今后研制新型、有效的PEDV疫苗奠定了基础。
In this study,the pig intestinal tract and its contents collected from a suspected PEDV infected in a pig farm in Henan were treated and inoculated into Vero cells and subjected to continuous in vitro subculture.The strain was successfully isolated and identified in vitro by S gene genetic evolution analysis,RT-PCR and indirect immunofluorescence assay,and named as PEDV HNLB.The strain was able to stably pass on Vero cells and produce typical cytopathic effects(CPEs)with a 6th generation virus titer of 104.3 TCID50/mL.The successful isolation of the HNLB strain laid the foundation for the development of a new and effective PEDV vaccine in the future.
