促分裂原活化的蛋白激酶与表皮生长因子受体在三阴性乳腺癌患者中的表达及临床意义

Expression and clinical significance of mitogen-activated protein kinase and epidermal growth factor receptor in patients with triple negative breast cancer

目的探讨促分裂原活化的蛋白激酶(MAPK)及表皮生长因子受体(EGFR)在三阴性乳腺癌(TNBC)患者中的表达及临床意义。方法选取200例TNBC患者的TNBC组织及其中80例患者的癌旁组织,采用免疫组织化学SP法检测标本中MAPK及EGFR蛋白的表达情况,分析二者的关系,以及TNBC组织中MAPK和EGFR表达与淋巴结转移、临床分期、复发转移及生存的关系。结果TNBC组织中MAPK和EGFR的阳性表达率均明显高于癌旁组织,差异均有统计学意义(P﹤0.01)。TNBC组织中MAPK与EGFR的表达呈正相关(r=0.494,P﹤0.01)。不同临床分期、有无淋巴结转移及有无复发转移的TNBC患者的TNBC组织中MAPK、EGFR蛋白表达情况比较,差异均有统计学意义(P﹤0.05)。MAPK、EGFR阳性表达患者的5年无瘤生存率均明显低于阴性表达患者,差异均有统计学意义(P﹤0.01)。结论MAPK与EGFR在TNBC中的表达呈正相关,并与肿瘤的侵袭、转移关系密切,是TNBC预后不良的生物学指标之一。

Objective To investigate the expression of mitogen-activated protein kinase(MAPK)and epidermal growth factor receptor(EGFR)in patients with triple negative breast cancer(TNBC)and their clinical significance.Method A total of 200 TNBC tissues and 80 tumor-adjacent tissues from patients with TNBC were selected.Immunohistochemical SP method was used to detect the expression levels of MAPK and EGFR,and the association between them was analyzed.The association between MAPK,EGFR expression in TNBC tissues and lymph node metastasis,clinical staging,recurrence,metastasis and survival were analyzed.Result The expression of MAPK and EGFR in TNBC tissues were higher than that in tumor-adjacent tissues with statistically significant difference(P<0.01).There was a positive correlation between the expression of MAPK and EGFR in TNBC tissues(r=0.494,P<0.01).The expression of EGFR and MAPK in TNBC tissues of TNBC patients with different clinical staging,lymph node metastasis and relapsed or metastasis were of statistically significant differences(P<0.05).The 5-year tumor-free survival rate in patients with positive expression of MAPK and EGFR were significant lower than those with negative expression with statistically significant difference(P<0.01).Conclusion MAPK is positive correlated with the EGFR in TNBC.The two indicators have a close association with tumor invasion and metastasis and act as biomarkers for poor prognosis of TNBC.
