摘要
目的目的探讨急性冠脉综合征(acute coronary syndrome,ACS)患者血清miR-27a、miR-34a表达及与冠脉病变程度的相关性分析。方法132例ACS患者根据患者临床资料分为急性心肌梗死(AMI)组69例、不稳定性心绞痛(UA)组63例,另选取健康受试者57例纳入对照组。应用SYBR Green实时荧光技术定量检测3组空腹血清中miR-27a及miR-34a的表达水平。比较冠状动脉支数与冠脉病变程度不同ACS患者miR-27a、miR-34a水平的差异及血清miR-27a、miR-34a水平与冠脉病变程度相关性。结果AMI组和UA组的miR-27a、miR-34a水平均明显高于对照组(P<0.05),且AMI组的miR-27a、miR-34a水平明显低于UA组(P<0.05);ACS患者中冠状动脉支数与血清miR-27a、miR-34a水平比较差异有统计学意义(P<0.05);冠状动脉病变程度中的轻度组与中度组miR-27a、miR-34a水平均明显低于重度组(P<0.05),中度组的miR-27a、miR-34a水平明显高于轻度组(r值分别为0.834、0.772、0.731、0.762,P<0.05);急性冠脉综合征患者血清miR-27a、miR-34a水平与Gensini积分均呈正相关(P<0.05)。结论急性冠脉综合征患者血清miR-27a、miR-34a表达与冠脉病变程度随Gensini评分增高而加重,呈正相关,可检测血清miR-27a、miR-34a对ACS患者进行危险分层及预后分析,用于辅助诊断ACS提供参考。
Objective To investigate the correlation between the expressions of serum miR27a,miR34a and the severity of coronary lesions in patients with acute coronary syndrome(ACS).Methods A total of patients with ACS were divided into acute myocardial infarction group(AMI group,n=69)and unstable angina group(UA group,n=63).And fifty-seven healthy examinees were enrolled in control group.SYBR Green real-time technique was used to detect the expression levels of miR27a and miR34a in fasting serum of the patients in all groups.The difference between the numbers of coronary vessels and the levels of miR-27a and miR-34a of the ACS patients with different severity of coronary lesions were compared and the correlation between the serum levels of miR-27a and miR-34a and the severity of coronary lesions were analyzed.Results The levels of miR27a and miR34a in AMI group and UA group were significantly higher than those in control group(P<0.05),and the levels of miR-27a and miR-34a in AMI group were significantly lower than those in UA group(P<0.05).Moreover,there was significant difference between the numbers of coronary vessels of ACS patients and the serum levels of miR-27a and miR-34a(P<0.05).Furthermore,the levels of miR-27a and miR-34a of the ACS patients,which declined in severe group,moderate group and mild group,were positively correlated with Gensini score(P<0.05).Conclusion The expression of the serum levels of miR-27a and miR-34a and the stenosis degree are positively correlated to Gensini score in ACS patients.So the detection of the serum levels can be used to analyze the risk stratification and prognosis of ACS patients.
