葶苈生脉方对慢性充血性心力衰竭大鼠肠功能障碍的调节作用

Regulation of Tingli Shengmai Prescription on Intestinal Dysfunction in Rats with Chronic Congestive Heart Failure

目的:研究葶苈生脉方对慢性充血性心力衰竭(CHF)大鼠肠功能障碍的调节作用,探讨其作用机制。方法:将50只雄性SD大鼠随机分为假手术组、模型组、对照组、葶苈生脉方低剂量组、高剂量组,每组10只。除假手术组外,其余4组手术造成腹主动脉狭窄,假手术组只分离腹主动脉,不结扎。5组均于手术4周后开始给药(水),每日1次定量灌胃,连续灌胃8周。最后1次给药后,收集各组尿液及血液标本,检测血清炎性因子[肿瘤坏死因子-α(TNF-α)、白细胞介素-4(IL-4)、白细胞介素-8(IL-8)],血浆D-乳酸、二胺氧化酶(DAO),尿乳果糖/甘露醇(L/M)。结果:与假手术组比较,模型组血清炎性因子TNF-ɑ、IL-4、IL-8,血浆D-乳酸、DAO,L/M均显著升高(P<0.01);与模型组比较,葶苈生脉方低、高剂量组血清炎性因子TNF-ɑ、IL-4、IL-8,血浆D-乳酸、DAO,L/M均明显降低(P<0.01)。结论:葶苈生脉方可有效改善CHF大鼠肠功能障碍,其作用机制可能与抑制炎性细胞因子释放,降低肠黏膜通透性损伤有关。

Objective:To study the regulating effect of Tingli Shengmai Prescription on intestinal dysfunction in rats with chronic congestive heart failure(CHF)and explore its mechanism.Methods:Fifty male SD rats were randomly divided into sham-operated group,model group,control group,low-dose group and high-dose group with 10 rats in each group.Except for the sham-operated group,the other four groups were operated on to cause abdominal aorta stenosis.The abdominal aorta was only separated in the sham-operated group without ligation.After 4 weeks of operation,all the five groups were administered(water)orally once a day for 8 weeks.After the last administration,urine and blood samples were collected and serum inflammatory factors(TNF-α,IL-4,IL-8),plasma D-lactic acid,diamine oxidase(DAO),lactulose/mannitol(L/M)were detected.Results:Compared with sham-operated group,TNF-α,IL-4,IL-8,plasma D-lactic acid,DAO,L/M in model group were significantly increased(P<0.01);compared with model group,TNF-α,IL-4,IL-8,plasma D-lactic acid,DAO,L/M in low and high dose groups were significantly decreased(P<0.01).Conclusion:Tingli Shengmai Prescription can effectively improve intestinal dysfunction in CHF rats,and its mechanism may be related to inhibiting the release of inflammatory cytokines and reducing intestinal permeability damage.
