大剂量生白术配伍枳实对慢传输型便秘大鼠结肠5-HT3R、5-HT4R表达的影响 被引量:44

Effects of High-dose Raw Rhizoma Atractylodis Macrocephala Combined with Fructus Aurantii Immaturus on the Expression of 5-HT3R and 5-HT4R in Colon of Rats with Slow Transit Constipation

摘要 目的研究大剂量生白术(70 g)配伍枳实(30 g)对慢传输型便秘(slow transit constipation,STC)大鼠结肠组织中5-HT3R、5-HT4R表达的影响。方法采用大黄酸灌胃法建立STC大鼠模型。将造模成功大鼠27只随机分为模型对照组、普芦卡必利组和枳术组,每组9只,另设10只为健康对照组。健康对照组、模型对照组予自然喂养,普芦卡必利组予0.018 mg/100 g(1 mL/100 g)普芦卡必利悬液灌胃,枳术组予0.9 mL/100 g枳术煎剂灌胃,连续给药4周。炭末推进率检测大鼠肠道传输功能,Real-time PCR及免疫组化检测大鼠结肠组织中5-HT3R、5-HT4R mRNA及蛋白的表达。结果与健康对照组比较,模型对照组大鼠活性炭推进率降低(P<0.05),5-HT3R、5-HT4R mRNA及蛋白表达均降低(P<0.05);与模型对照组比较,普芦卡必利组和枳术组大鼠活性炭推进率增强(P<0.05),5-HT3R、5-HT4R mRNA及蛋白表达均升高(P<0.05);普芦卡必利组和枳术组比较,差异无统计学意义(P>0.05)。结论大剂量生白术配伍枳实可以上调STC大鼠结肠组织中5-HT3R、5-HT4R mRNA及蛋白的表达,促进肠道动力。 Objective To investigate the effect of high-dose raw Rhizoma atractylodis macrocephala combined with Fructus aurantii immaturus on the expression of 5-HT3 receptors and 5-HT4 receptors in colon tissue of slow transit constipation(STC)rats.Methods The STC rat model was established by gavage with rheum officinale.Twenty-seven rats with successful model establishment were randomly divided into the model group,the Prucalopride group,and the Rhizoma atractylodis macrocephala-Fructus aurantii immaturus(Zhizhu)group,9 in each group,and 10 SD rats were recruited as normal group.The normal group and the model group rats were given natural feeding.The Prucalopride group rats were intragastrically administered 0.018 mg/100 g(1 mL/100 g)Prucalopride suspension,and the Zhizhu group rats were given 0.9 mL/100 g of Zhizhu Decoction.Every group were continuously fed for 4 weeks.The intestinal transmission function of rats was detected by the promote rate of carbon powder.The expression of 5-HT3 and 5-HT4 receptors mRNA and protein were detected by Real-time PCR and immunohistochemistry,respectively.Results Compared with the normal group,the promote rate of carbon powder,the express of 5-HT3 receptors and 5-HT4 receptors mRNA and protein decreased in the model group(all P<0.05).Compared with the model group,the promote rate of carbon powder was enhanced(P<0.05),as well as the expression of 5-HT3 receptors and 5-HT4 receptors mRNA and protein increased in the Prucalopride group and the Zhizhu group(P<0.05).There were no statistical difference between the Prucalopride group and the Zhizhu group(P>0.05).ConclusionHigh-dose raw Rhizoma atractylodis macrocephala combined with Fructus aurantii immaturus could increase the expression of the 5-HT3 and 5-HT4 receptors mRNA and protein in STC rat colon tissues,and promote the intestinal motility.
