经皮穴位电刺激对高龄妇女卵细胞质量及β-catenin的影响 被引量:8

Effects of percutaneous acupoint electrical stimulation on oocyte quality andβ-catenin in older women

摘要 目的:通过研究在体外受精(IVF)周期中经皮穴位电刺激对高龄妇女卵细胞质量及β-catenin的影响,探讨经皮穴位电刺激影响高龄妇女卵细胞质量的机制。方法:将88例行体外受精-胚胎移植(IVF-ET)的高龄妇女随机分为两组,试验组(经皮穴位电刺激合西药治疗)和对照组(伪针合西药治疗),每组各44例。分别观察两组患者治疗前后证候积分及积分差值、促性腺激素(Gn)总量、取卵数、单卵雌激素水平、受精率、卵裂率、优质胚胎率、妊娠率、临床妊娠率,取卵日卵巢颗粒细胞Wnt通路中β-catenin蛋白水平的表达差异。结果:与对照组比较,试验组单卵雌激素水平提高(P<0.05),优胚率、妊娠率及临床妊娠率显著提高(P<0.05),颗粒细胞β-catenin的表达水平显著上调(P<0.05)。结论:经皮穴位电刺激提高高龄妇女卵细胞质量的机制与调控卵巢颗粒细胞Wnt通路,上调通路中关键因子β-catenin蛋白的表达,促进颗粒细胞增殖、改善卵巢微环境有关,其作用机制尚需进一步探讨。 Objective:Through the research in in vitro fertilization(IVF)cycle,skin acupoint stimulation for older women oocyte quality and influence ofβ-catenin,to study the effects of percutaneous acupoint stimulation mechanism of the egg quality of older women.Methods:Eighty-eight IVF-ET women were randomly divided into 2 groups,experimental group(transcutaneous point electric stimulation combined western medicine group)and control group(pseudo needle western medicine group)and 44 cases in each group.The scores were observed before and after treatment in both groups and integral differential,gonadotropin(Gn),total oocyte number,single oocyte estrogen levels,fertilization rate,cleavage rate,high quality embryo rate and pregnancy rate,clinical pregnancy rate,take oocytes,ovarian granulosa cells Wnt pathways in beta catenin protein expression levels of the difference.Results:Compared with control group,the single oocyte estrogen levels in the experimental group increase significantly(P<0.05),the optimal embryo rate,pregnancy rate and clinical pregnancy rate increased significantly(P<0.05),and the granulosa cellsβ-catenin expression level raised obviously,statistically significant difference(P<0.05).Conclusion:Percutaneous acupoint stimulation mechanism to improve the quality of older women’s oocytes and regulation of ovarian granulosa cells Wnt pathways,raised pathways of key factor beta-catenin protein expression,promote and improve ovarian granulosa cell proliferation micro environment,the mechanism still needs further discussion.
