刺山柑体外抗肿瘤活性提取物的筛选研究 被引量:4

摘要 目的:分离萃取得到刺山柑的五个提取物,筛选对刺山柑提取物最敏感的人肿瘤细胞系和对人肿瘤细胞抑制作用最强的活性提取物。方法:以85%乙醇提取刺山柑药材,分别用等体积的石油醚、氯仿、乙酸乙酯萃取得石油醚提取物、氯仿提取物和乙酸乙酯提取物,剩余的水溶液部分减压浓缩后干燥得水溶性提取物;CCK-8法测定有效提取物的体外抑制作用。结果:刺山柑五个提取物对人非小细胞肺癌细胞A549、人乳腺癌细胞MCF-7、人卵巢癌细胞SK-OV-3、人前列腺癌细胞PC-3、人肝癌细胞Hep-G2、人结肠癌细胞HT-29这六种常见的人肿瘤细胞均有抑制作用,其半抑制浓度(IC50值)范围为:350.503~1 047.614μg·mL^-1。其中以石油醚提取物对人卵巢癌细胞SK-OV-3效果最好,其对应的最大抑制率和IC50值分别为95.801%和350.503μg·mL^-1。结论:人卵巢癌细胞SK-OV-3对刺山柑石油醚提取物最为敏感。 Objective:To select the most sensitive human tumor cell lines to the effective extracts of capparis spinosa L.and the most active site which has the strongest inhibitory effect on human tumor cells from the five effective extracts of capparis spinosa L..Methods:The capparis spinosa L.were collected with 85%ethanol,and then the crude extract were extracted with equal volume of petroleum ether,chloroform and ethyl acetate to obtain the extracts of petroleum ether,chloroform,and ethyl acetate.The remaining aqueous solution was concentrated under reduced pressure and dried to get a water-soluble portion.The inhibitory effect of the effective extract was determined by CCK-8 method.Results:Five effective extracts of capparis spinosa L.had the antitumor effect to human non-small cell lung cancer cells A549,human breast cancer cells MCF-7,human ovarian cancer cells SKOV-3,human prostate cancer cells PC-3,human hepatoma cells Hep-G2,human colon cancer cell line HT-29,and the 50%inhibiting concentration(IC50 value)ranged from 350.503 to 1 047.614μg·mL^-1.The extract of petroleum ether had the best effect on human ovarian cancer cell SK-OV-3,and the corresponding maximum inhibition rate and IC50 value were 95.801%and 350.503μg·mL^-1.Conclusion:The experimental results showed human ovarian cancer cell line SK-OV-3 was most sensitive to petroleum ether extract parts of capparis spinosa L..
