摘要
目的探讨GS环保试剂代替二甲苯应用于非小细胞肺癌表皮生长因子受体(epidermal growth factor receptor,EGFR)基因突变检测的效果。方法选取16例确诊为肺腺癌的标本,同一蜡块分别用GS环保脱蜡液、二甲苯进行脱蜡,提取DNA,应用实时荧光定量PCR法检测EGFR基因突变状态。结果GS环保试剂与二甲苯脱蜡同一石蜡标本,GS环保试剂对石蜡切片脱蜡效果良好,标本DNA含量、质量差异无统计学意义(P>0.05),且均符合实时荧光定量PCR要求,且应用实时荧光定量PCR法检测EGFR基因突变状态,其基因突变类型与使用二甲苯脱蜡的检测结果完全一致(符合率100%)。结论GS环保试剂可代替二甲苯应用于非小细胞肺癌EGFR基因突变检测,并符合实验室环保的要求。
Objective To explore the effect of an environmental reagent GS for the detection of epidermal growth factor receptor(EGFR)gene mutation in non-small cell lung cancer instead of xylene.Methods Sixteen specimens of lung adenocarcinoma were selected.The same wax block was dewaxed with environmental reagent GS and xylene respectively,and then DNAs were extracted from the specimens.Real-time fluorescence quantitative PCR was performed to detect the mutation state of EGFR gene.Results The environmental reagent GS had a good dewaxing effect on paraffin-embedded tissue sections compared with xylene.There was no statistical difference in the content and quality of sample DNA between environmental reagent GS and xylene dewaxing(P>0.05).And the content and quality of sample DNA were in accordance with the requirement of real-time fluorescence quantitative PCR.In addition,the detection of the mutation state of EGFR gene was performed using real-time fluorescence quantitative PCR.The types of gene mutation using environmental reagent dewaxing were completely consistent with the results using xylene dewaxing(100%coincidence rate).Conclusion The green reagent dewaxing agent GS can be used in the detection of EGFR gene mutation in non-small cell lung cancer instead of xylene,and it is in accordance with the requirement of the laboratory environmental protection.
