HPLC法测定知柏地黄丸中毛蕊花糖苷的含量

Determination on the Content of Verbascoside in Zhibo dihuang Pills by HPLC

目的 建立测定知柏地黄丸中毛蕊花糖苷含量的方法。方法 采用高效液相色谱法,色谱柱为Agilent C18柱(4.6 mm×250 mm×5 μm),流动相为乙腈-0.1%乙酸溶液(13∶87,V/V),流速为1.0 mL/min,柱温为33 ℃,检测波长为334 nm,进样量为10 μL。结果 毛蕊花糖苷检测质量浓度在5.2×10^-3~208×10^-3 mg/mL浓度范围内线性关系良好(r=0.9998);精密度、稳定性、重复性试验的RSD≤1.37%;平均加样回收率为97.40%,RSD=1.28%(n=6)。结论 该法简便、准确、灵敏度高、重复性好,适用于测定知柏地黄丸中毛蕊花糖苷的含量。

Objective To establish a method for the content determination of verbascoside in zhibai dihuang Pills by RP-HPLC.Methods HPLC method was adopted.The determination was performed on Agilent C18 column(4.6 mm×250 mm×5 m)with mobile phase consisted of acetonitrile-0.1%ethylic acid solution(13:87,V/V)at the flow rate of 1.0 ml/min.The column temperature was 33℃,and detection wavelength was set at 334 nm.The sample size was 10μl.Results The linear range of verbascoside was 5.2×10^-3~208×10^-3 mg/ml(r=0.9998)with an average recovery of 97.40%(RSD=1.28%,n=6).RSDs of precision,stability and reproducibility tests were lower than 1.21%.Conclusion The method is simple,accurate,sensitive and reproducible,which can be used for the content determination of verbascoside acid in Zhibo dihuang pills.
