摘要
目的探讨外周血中微小RNA-34a(miR-34a)在乳腺癌组织中的表达及其临床病理学意义。方法选取乳腺癌组织标本80例(癌组织)、40例癌旁组织标本(癌旁组织),应用RT-PCR技术检测两组标本中的miR-34a表达水平,采用原位杂交方法检测miR-34a的表达,分析不同miR-34a表达与乳腺癌患者临床病理学特征的关系。结果乳腺癌组织中的miR-34a表达水平显著的低于癌旁组织,差异具有统计学意义(P<0.05);乳腺癌组织中的miR-34a表达水平与患者的TNM分期、浸润深度、淋巴结转移具有显著的相关性(P<0.05);乳腺癌组织中的miR-34a表达水平与患者的年龄、肿瘤直径、分化程度、肿瘤类型、ER表达、PR表达的情况无显著关系(P>0.05);低表达miR-34a乳腺癌患者随访60个月生存21例(46.67%)、失访3例,高表达miR-34a乳腺癌患者生存20例(64.52%)、失访1例,两组生存率比较差异无统计学意义(χ2=2.354,P=0.125>0.05)。低表达miR-34a乳腺癌患者生存时间低于高表达miR-34a乳腺癌患者[Log Rank(Mantel-Cox)=3.928,P=0.047<0.05]。结论乳腺癌组织中miR-34a低表达,并且与患者TNM分期、浸润深度、淋巴结转移、不良预后有关。
Objective To investigate the expression of RNA-34a(miR-34a)in peripheral blood and its clinicopathological significance in breast cancer.Methods The expression of microRNA-34a in breast cancer tissues from 80 breast cancer patients,40 cases of paracancerous tissues were detected by RT-PCR and in situ hybridization.The relationship between the expression of microRNA-34a and clinicopathological characteristics of breast cancer patients was analyzed.Results The expression of miR-34a in breast cancer was significantly lower than that of the adjacent tissues,the difference was statistically significant(P<0.05);breast cancer tissues and the expression level of miR-34a in patients were significantly correlated with TNM stage,depth of invasion,and lymph node metastasis(P<0.05);breast cancer tissue miR-34a levels in patients had no significant relationship with the age,the diameter of tumor,differentiation,tumor type,ER expression,PR expression(P>0.05);the follow-up of miR-34a breast cancer patients with low expression 60 months survival was 21 cases(46.67%),3 cases were lost,the high expression of miR-34a in breast cancer survival in 20 cases(64.52%),1 cases were lost,two group survival rate had no significant difference(χ2=2.354,P=0.125>0.05).The survival time of breast cancer patients with low expression of miR-34a was lower than that with high expression of miR-34a(Log Rank(Mantel-Cox)=3.928,P=0.047<0.05).Conclusion Low expression of miR-34a in breast cancer tissues is associated with TNM stage,depth of invasion,lymph node metastasis,and poor prognosis.
