彭波半细毛羊BMP15和BMPR-IB基因突变与其产羔性的相关分析 被引量:8

Correlation Between BMP15 and BMPR-IB Gene Mutations in Pengbo Semi-fine Wool Sheep and Their Lambing

摘要 BMP15基因(bone morphogenetic protein 15,BMP15)和BMPR-IB基因(bone morphogenetic protein 1beta,BMPR-IB)是影响绵羊排卵率和产羔数的主效基因,对绵羊多羔性状有明显加性效应,可作为检测绵羊繁殖力的主要辅助标记.利用操作简便、灵敏度高等特点的高分辨率熔解曲线分析(High resolution melting curve,HRM),并结合测序技术,对63只彭波半细毛羊(产单羔羊56只、产双羔羊7只)BMP15和BMPR-IB基因的常见突变区域进行突变分析.结果表明:所有样本BMP15基因第718位、第747位、第755位、第871位和第896位,BMPR-IB基因233位和746位均未见DNA突变;仅23个样本的BMPR-IB基因1 078位发现T到C的杂合性突变.但经相关性检验分析,发现该杂合性突变与彭波半细毛羊的双羔性无关. Bone morphogenetic protein 15 gene(BMP15)and bone morphogenetic protein 1 beta(BMPRIB)are the major genes that could affect ovulation rate and litters′number of sheep.Both of the two genes have obvious additive effects on sheep traits.Hence,BMP15 and BMPR-IB can be used as assistant markers for detecting the fecundity of sheep.In the present study,high-resolution melting(HRM)method,combining with DNA sequencing,was used to analyze 63 Pengbo semi-fine wool sheep samples(56 singleton lambs and 7 twins lambs)for mutations in BMP15 and BMPR-IB genes.The results indicated that no mutations in suspected mutational region in BMP15 gene were detected and only one heterozygous mutation of T to C was found at position 1078 of BMPR-IB genein 23 samples.However,the correlation test showed that the detect?ed heterozygous mutation had nothing to do with the double lambing of Pengbo semi-fine wool sheep.
